چکیده

The almost unique localization of hydroxyproline in collagen and the availability of methods for the assay of the imino acid offer a convenient approach to studies on this protein. In the experiments described here, L-proline-Cl4 was injected into embryonated eggs, and the specific activities of protein-bound proline and hydroxyprolinei in various subcellular fractions of the embryo were determined. Protein-bound hydroxyproline with the highest specific activity appeared in the microsomal fraction, which suggests that collagen is synthesized by mechanisms imilar to those which have been implicated in the synthesis of other proteins (1).

Effect of Adenosine Agonists on the Proliferation and Differentiation of Chick Embryo Fibroblasts in Three Dimensional Reconstituted Tissue Constructs

Previous studies indicate that organ fibroblasts play an important role in wound healing, collagen production, remodeling processes and pathogenesis of progressive heart, lung, renal and hepatic fibrotic diseases. Several studies suggest a possible inhibitory role for adenosine in the regulation of fibroblast proliferation. The effect of adenosine A2 agonists on proliferation and differentiatio...

A Chick Embryo in-Vitro Model of Knee Morphogenesis

Background: In this feasibility study, a mechanically loaded in-vitro tissue culture model of joint morphogenesis using the isolated lower extremity of the 8 day old chick embryo was developed to assess the effects of mechanical loading on joint morphogenesis. Methods: The developed in-vitro system allows controlled flexion and extension of the chick embryonic knee with a range of motion of ...

Study of Basement Membrane Type IV Collagen Appearance in the Brain Choroids Plexus of Mouse Fetuses

Introduction & Objective: The brain choroids plexus (BCP) plays an important role in the cerebrospinal fluid (CSF) production, but its characterization is still incomplete. Collagen type IV, is one of the most important proteins of basement membrane (BM) and extracellular matrix (ECM) of BCP. In the present study we investigated the differential period of type IV collagen in basement membrane...

pH-dependent function, purification, and intracellular location of a major collagen-binding glycoprotein

A major collagen-binding heat shock protein of molecular mass 47,000 D was found to bind to collagen by a pH-dependent interaction; binding was abolished at pH 6.3. Native 47-kD protein could therefore be purified from chick embryo homogenates in milligram quantities by gelatin-affinity chromatography and gentle acidic elution. Rat monoclonal and rabbit polyclonal antibodies were generated agai...

Collagen Metabolism in the Normal and Lathyritic Chick

1. Radioisotope incorporation studies of normal and lathyritic chick embryo bone collagen do not demonstrate any interference by lathyrism with collagen synthesis or fibril formation. 2. The results indicate that a portion of the extractable collagen from lathyritic chick embryo bone represents newly synthesized protein. Evidence from a double labeling experiment and from analysis of isotope fl...
