چکیده

There is increasing evidence for cell-to-cell interactions in immune response (1, 2). While the interaction between B and T lymphocytes has been studied by many investigators both in vivo and in vitro, there is a paucity in the literature concerning subpopulations of T cells and their interactions. The nature of T-Tcell interactions is considered an important key in resolving the regulatiry mechanisms of cell-mediated immune responses (CMI). 1 Cantor et al. (3, 4) first reported the cooperation of two T-cell subpopulations, termed precursor of effector cells and amplifier cells, in the development of graft-vs.-host reactions (GVH) in mice. Suppressive actions o f t cells were also demonstrated in GVH by Gershon et al. (5, 6) and by Hardin et al. (7). T-T-cell cooperations and suppression in vitro have been observed in mixed lymphocyte reactions (MLR) (8-11) and in cytotoxic allograft responses (12-15). Delayed hypersensitivity (DH) is an important phenomenon manifested by T-cell activity, yet few papers have discussed such activity. A decline in immunocompetence with respect to both cell-mediated and humoral immunity is considered an age-related phenomenon in mammalians and this decline appears to be linked to involution of the thymus (16). In the present study using mice, we found with age, an increasing reactiveness in DH to methylated human serum albumin (MHSA). After reaching peak responses at around 10-15 mo of age, the animals showed a gradual reduction in the degree of the response. In a preceding p a p e r , 2 w e reported that foot pad reaction (FPR) as a manifestation of DH to MHSA is a phenomenon with high T-cell function dependence, and that the grade of foot pad swelling was limited by the sensitized T-cell population. The discrepancy observed in involution of the thymus and

Regulatory T Cells and Myeloid-Derived Suppressor Cells in Patients with Peptic Ulcer and Gastric Cancer

Background: Regulatory T Cells (Tregs) and Myeloid-Derived Suppressor Cells (MDSCs) are two main regulatory cells modulating the immune responses in inflammation and cancer. Objective: To investigate and compare Tregs and MDSCs in peptic ulcer and gastric cancer. Methods: Patients with dyspepsia were selected and divided into three groups of non-ulcer dyspepsia (NUD, n=22), peptic ulcer disease...

Multiple Low Doses of 5-Fluorouracil Diminishes Immunosuppression by Myeloid Derived Suppressor Cells in Murine Melanoma Model

Background: Melanoma progression and metastasis is suggested to be mediated by increased accumulation of myeloid derived suppressor cells. Various chemotherapeutic drugs such as 5-Fluorouracil in single low concentration have the capacity, at least in part, to reverse tumor progression by reducing myeloid derived suppressor cellsmediated immunosuppression. Objective: To assess whether multiple ...

Altered Suppressor Function of Regulatory T Cells in Type 1 Diabetes

Background: Type 1 diabetes (T1D) is a T cell mediated autoimmune disease targeting the insulin-producing β cells within pancreatic islets. Autoimmune diseases may develop as a consequence of altered balance between regulatory (Tregs) and autoreactive T cells. Objectives: To evaluate Treg cells frequency and suppressive function in the peripheral blood of newly diagnosed T1D patients in compari...

Cyclophosphamide eliminates suppressor T cells in age-associated central regulation of delayed hypersensitivity in mice

Effect of treatment of mice with cyclophosphamide (CY) on the delayed hypersensitivity (DH) response was investigated in C57BL/6 mice. DH to methylated human serum albumin (MHSA) could be enhanced with CY in young mice but not in aged ones. DH enhancement with CY appeared to be due to elimination of suppressor T cells involved in DH. Effector T cells were also sensitive to CY, the damaging effe...

Delayed-type hypersensitivity to influenza virus. Induction of antigen- specific suppressor T cells for delayed-type hypersensitivity to hemagglutinin during influenza virus infection in mice

Mice infected with A/England/939/69 X A/Puerto Rico/8/34 (Rec 31) influenza virus by aerosol develop significantly lower levels of delayed-type hypersensitivity (DTH) to A/Hong Kong/1/68 X A/Puerto Rico/8/34/ (X31) virus compared to uninfected mice. The suppression of DTH to the hemagglutinin appears to be mediated by suppressor T cells which carry Lyt-1 membrane antigen marker, and not by sy s...

Mechanisms of regulation of cell-mediated immunity. III. The characterization of azobenzenearsonate-specific suppressor T-cell- derived-suppressor factors

Delayed type hypersensitivity to the hapten azobenzenearsonate (ABA) can be induced and suppressed by the administration of hapten-coupled syngeneic spleen cells by the appropriate route. Suppressor T cells stimulated by the intravenous administration of ABA-coupled spleen cells have been shown to produce a discrete subcellular factor(s) which is capable of suppressing delayed type hypersensiti...
