The cell-bound penicillinase of Bacillus cereus.

چکیده
Only 30-50 yo of the cell-bound penicillinase of Bacillus cereus NRRL 569 is neutralized by antiserum prepared against the exo-penicillinase. The unneutralizable fraction is not decreased by cell disintegration which liberates a proportion of the cell-bound enzyme into solution. Absence of neutralization cannot therefore be explained by the existence of a mechanical barrier which prevents access of antibody; it has in fact been shown to be due to the presence of another type of penicillinase, not liberated from the cells, which is enzymically similar to, but immunologically and physico-chemically distinct from, the exo-enzyme. This cellbound penicillinase has been separated from the exo-enzyme by fractional precipitation with ammonium sulphate. It is inducible by penicillin, like the exo-enzyme, and is present in cells of the constitutive mutant strain, 569/H.

The changes in wall structure of a penicillinase micro-constitutive strain of Bacillus cereus (569/H/24), on exposure to penicillin, and after its removal by addition of penicillinase, have suggested the following model for the growth of the walls of these cylindrical cells. Longitudinal extension is by addition of material to a large and continuously increasing number of growing points uniform...

متن کاملThe effect of urea and guanidine hydrochloride on activity and optical rotation of penicillinase.
The penicillinase of Bacillus cereus exists as an exoenzyme, called cw-penicillinase, and as an immunologically distinct cellbound enzyme, designated as y-penicillinase (1). Properties such as substrate specificity, Michaelis constant, and sedimentation rate were found to be practically identical, and the only other ways in which the cell-bound y-penicillinase differed from LYpenicillinase were...
