Utilization of Fixed Nitrogen by Azotobacter and Influence on Nitrogen Fixation.

چکیده
The influence of fixed nitrogen on Azotobacter both with respect to utilization and inhibition of nitrogen fixation has been studied by a number of investigators. The results have not always been conclusive because of the relatively poor growths of cultures obtained in culture media now known to have been deficient in one or more of the elements essential for nitrogen fixation or growth, particularly calcium, iron or molvbdenum. Inadequate analyses of the fixed nitrogen consumed by the organisms, which, with few exceptions, were cultured in the presence of nitrogen gas, also leads to uncertainty. The results reported in this paper were obtained by testing a variety of nitrogenous compounds on several strains of Azotobacter in a medium capable of giving growth equivalent to 20 to 30 mg. nitrogen per 100 ml. in a few days. The experiments wvere carried out chiefly in the absence of nitrogen gas in atmospheres containing one-fifth of an atmosphere of oxygen only or with hydrogen added to bring the total pressure to one atmosphere. Under these conditions any visible growth is evidence of utilization of the fixed nitrogen added uncomplicated by any simultaneous assimilation of nitrogen gas. Recent investigations in this laboratory, as well as those of Wyss et al. (1941), have shown that whereas hydrogen inhibits nitrogen fixation, it has no significant effect on the growth of Azotobacter supplied with the usual quantities of readily available fixed nitrogen. In addition to these experiments made in the absence of nitrogen gas a few cultures were grown in the presence of air, with several nitrogen compounds added to the medium in order to test the relative utilization of free and fixed nitrogen.

The Utilization of Nitrogen in Hydroxylamine and Oximes by Azotobacter vinelandii.
Recent studies have provided several types of evidence supporting the view that ammonia is the key intermediate in nitrogen fixation by Azotobacter. An alternative hypothesis based on hydroxylamine depends almost entirely on the detection of traces of oximes in cultures fixing nitrogen. Although the two hypotheses are not mutually exclusive (Burris and Wilson, 1945), hydroxylamine remains an im...

Investigations with the isotopic tracer N16 have furnished definite evidence that ammonia is the key intermediate in nitrogen fixation by Azotobacter vinelundii (1). Direct isolation of the intermediate, however, has not been possible, apparently because ammonia is used so rapidly by this bacterium that it does not accumulate. In a study of the comparative biochemistry of nitrogen fixation, we ...
متن کاملThe utilization of nitrogen in various compounds by Clostridium pasteurianum.
In biological nitrogen fixation the key intermediate compound, defined as the end product of the fixation reaction and the initial reactant of assimilation, is believed to be either ammonia or hydroxylamine. Burris and Wilson (1945) noted that these two possibilities need not be mutually exclusive but may occur simultaneously, depending on the concentrations of the carbon compounds serving as a...
متن کاملThe Influence of Roentgen Rays upon the Nitrogen Fixation by Azotobacter
The influence of graded x-ray doses upon nitrogen fixation and respiration by Azotobacter was studied by means of the Warburg method. It was found that nitrogen fixation decreases approximately linearly with increasing x-ray doses. Respiration in contrast is affected only indirectly through some inhibition of cell multiplication. Small doses of x-ray often produce a slight and transient increas...
متن کاملMutants of azotobacter that do not fix nitrogen.
Stable strains of mutants of Azotobacter that do not fix nitrogen are difficult to obtain by traditional procedures (Karlsson and Barker, 1948; Lindstrom, 1948). Stumbo and Gainey (1938) demonstrated that culturing Azotobacter on a high nitrate medium resulted in strains that failed to fix nitrogen even after continued transfer in the absence of fixed nitrogen. However, since these strains were...
