Tyrosine kinase receptor B isoforms alter APP and BACE1 endogenous levels independently of BDNF

چکیده
Brain derived neurotrophic factor (BDNF) levels and signaling via the tyrosine receptor kinase B (TrkB) have been shown to be altered in Alzheimer’s Disease. In addition, it has been reported that the isoforms of TrkB can differentially affect metabolism of amyloid precursor protein (APP). Conversely, A, a neurotoxic cleavage product of APP, has been shown to impair TrkB/ BDNF signaling. Therefore, we investigated whether the changes observed in APP metabolism were due to the isoform-specific effects of TrkB on either APP expression, and/or on the expression and activity of ADAM10 and BACE1. Since BDNF levels are decreased in AD, we focused on BDNF independent effects of the TrkB isoforms. We found that TrkB FL increases endogenous APP levels in both HEK293 and SH-SY5Y naïve cells. We did not find an increase in ADAM10 activity in HEK293 cells, but an increase in BACE1 levels. Additionally, we have found that TrkB FL is able to increase NFAT3 mediated transcriptional activity and we suggest that this causes transcriptional activation of the BACE1 promoter.

Effect of Endurance Training on Brain Derived Neurotrophic Factor (BDNF) and Tyrosine Kinase B (Trkb) Level in Hippocampus of Ischemic Induced Male Rats
Introduction: Brain derived neurotrophic factor (BDNF) have neuroprotective effect through binding with tyrosine kinase B (TrkB). Thus the Aim of the present study was to investigate the effects of eight weeks endurance training on BDNF and TrkB levels in the hippocampus of ischemic induced male rats. Methods: 40 Male wistar rats (12 weeks old and 228.19±21.18g) were divided into four groups, i...
متن کاملTrkB Isoforms Differentially Affect AICD Production through Their Intracellular Functional Domains
We report that NTRK2, the gene encoding for the TrkB receptor, can regulate APP metabolism, specifically AICD levels. Using the human neuroblastoma cell line SH-SY5Y, we characterized the effect of three TrkB isoforms (FL, SHC, T) on APP metabolism by knockdown and overexpression. We found that TrkB FL increases AICD-mediated transcription and APP levels while it decreases sAPP levels. These ef...
متن کاملInduction of tyrosine kinase receptor b by retinoic acid allows brain-derived neurotrophic factor-induced amyloid precursor protein gene expression in human SH-SY5Y neuroblastoma cells.
Retinoic acid (RA) is a potent regulator of morphogenesis, growth and cell differentiation. Incubation with RA causes arrest of proliferation and neurite extension in SH-SY5Y cells, a neuroblastoma cell line of human origin. In these cells, RA regulates the expression of the beta-amyloid precursor protein. The retinoid increases the levels of intracellular and secreted forms of APP (amyloid pre...
