disease and/or platelet activation, and may not participate directly in pathogenesis. We conclude that raised sP-selectin indicates a poor prognosis among patients with peripheral artery disease. The wide range of values means that this marker alone is unlikely to be of value in defining the risk of disease progression in any individual subject. However, it may be useful as part of a panel of markers that may include those suggestive of a pro-thrombotic capacity and endothelial cell dysfunction. If raised sP-selectin is truly a marker of platelet activation (8, 9), this may provide a new rationale for developing and using therapy aimed at the platelet. This preliminary finding may be applicable to other forms of atherosclerosis (e.g. stroke), and warrants further attention in larger groups of patients with more diverse disease(s).

Modulation by vitamin B6 of glucocorticoid receptor-mediated gene expression requires transcription factors in addition to the glucocorticoid receptor.
We have investigated the mechanism by which vitamin B6 acts to modulate steroid hormone-mediated gene expression. We show that the level of glucocorticoid-induced gene expression from simple promoters, containing only hormone response elements and a TATA sequence, was not affected by alterations in intracellular vitamin B6 concentration. However, modulation of hormone-induced gene expression wa...
متن کاملRisk factors for adrenal crisis in patients with adrenal insufficiency.
Patients with adrenal insufficiency have a life-threatening risk of adrenal crisis, thus preventing adrenal crisis is an important clinical issue. In order to clarify the risk factors for adrenal crisis, the medical records of 137 patients with established adrenal insufficiency were retrospectively investigated. The explanatory variables analyzed were gender, etiology of hypoadrenalism, class o...
متن کاملGenetic and clinical features of p450 oxidoreductase deficiency.
P450 oxidoreductase (POR) deficiency is an autosomal recessive disorder of steroidogenesis with multiple clinical manifestations. POR is the electron donor for all microsomal P450 enzymes, including the three steroidogenic enzymes P450c17 (17alpha-hydroxylase/17,20-lyase), P450c21 (21-hydroxylase), and P450aro (aromatase). Since the first description of POR mutations in 2004, about 50 patients ...
متن کاملSuppression of steroidogenesis and activator protein-1 transcription factor activity in rat adrenals by vitamin E deficiency-induced chronic oxidative stress.
Excessive oxidative stress and associated macromolecular damage are considered to be key features of aging, and appear to contribute to the age-related decline in steroid hormone production in adrenal and testicular Leydig cells. The current studies were initiated to examine the potential mechanism by which excessive oxidative stress during aging attenuates the functional expression of the oxid...
متن کاملVitamin B6 modulates transcriptional activation by multiple members of the steroid hormone receptor superfamily.
Recent studies have shown that vitamin B6 modulates transcriptional activation by the human glucocorticoid receptor in HeLa S3 cells. We have now examined the possibility that vitamin B6 might similarly influence transcriptional activation by the glucocorticoid receptor in other cell types, as well as gene expression mediated by other members of the steroid hormone receptor superfamily. We show...
