چکیده

31 Transcranial magnetic stimulation (TMS) can provide an index of intracortical 32 excitability/inhibition balance. However, the neurochemical substrate of these measures remains 33 unclear. Pharmacological studies suggest the involvement of GABAA and GABAB receptors in 34 TMS protocols aimed at measuring intracortical inhibition, but this link remains inferential. 35 Proton magnetic resonance spectroscopy (H-MRS) permits measurement of GABA and 36 glutamate + glutamine (Glx) concentrations in the human brain, and might help in the direct 37 empirical assessment of the relationship between TMS inhibitory measures and neurotransmitter 38 concentrations. In the present study, MRS-derived relative concentrations of GABA and Glx 39 measured in the left M1 of healthy participants were correlated with TMS measures of 40 intracortical inhibition. Glx levels were found to correlate positively with TMS-induced silent 41 period duration whereas no correlation was found between GABA concentration and TMS 42 measures. The present data demonstrate that specific TMS measures of intracortical inhibition are 43 linked to shifts in cortical Glx, rather than GABA neurotransmitter levels. Glutamate might 44 specificaly interact with GABAB receptors, where higher levels of MRS-derived Glx 45 concentrations seem to be linked to higher levels of receptor activity. 46 47

Comparing GABA-dependent physiological measures of inhibition with proton magnetic resonance spectroscopy measurement of GABA using ultra-high-field MRI

Imbalances in glutamatergic (excitatory) and GABA (inhibitory) signalling within key brain networks are thought to underlie many brain and mental health disorders, and for this reason there is considerable interest in investigating how individual variability in localised concentrations of these molecules relate to brain disorders. Magnetic resonance spectroscopy (MRS) provides a reliable means ...

Relationship between physiological measures of excitability and levels of glutamate and GABA in the human motor cortex

Magnetic resonance spectroscopy (MRS) allows measurement of neurotransmitter concentrations within a region of interest in the brain. Inter-individual variation in MRS-measured GABA levels have been related to variation in task performance in a number of regions. However, it is not clear how MRS-assessed measures of GABA relate to cortical excitability or GABAergic synaptic activity. We therefo...

An Exploratory Study of Spectroscopic Glutamatergic Correlates of Cortical Excitability in Depressed Adolescents

Introduction: Transcranial magnetic stimulation (TMS) research has suggested dysfunction in cortical glutamatergic systems in adolescent depression, while proton magnetic resonance spectroscopy (1H-MRS) studies have demonstrated deficits in concentrations of glutamatergic metabolites in depressed individuals in several cortical regions, including the anterior cingulate cortex (ACC). However, fe...

Neurochemical Effects of Theta Burst Stimulation as Assessed by Magnetic Resonance Spectroscopy

Continuous theta burst stimulation (cTBS) is a novel transcranial stimulation technique that causes significant inhibition of synaptic transmission for <or=1 h when applied over the primary motor cortex (M1) in humans. Here we use magnetic resonance spectroscopy to define mechanisms mediating this inhibition by noninvasively measuring local changes in the cortical concentrations of gamma-aminob...
