PGJ2, a new antiviral prostaglandin: inhibition of Sendai virus replication and alteration of virus protein synthesis.

چکیده
Prostaglandin J2 (PGJ2) was found to suppress dramatically Sendai virus replication in African green monkey kidney cells in culture. PGJ2 was not toxic at the active dose to uninfected cells and did not significantly inhibit macromolecular synthesis, but it specifically stimulated the synthesis of a polypeptide of 74,000 mol. wt. In Sendai virus-infected cells, PGJ2 partially inhibited virus protein synthesis and caused an alteration in the mobility of the virus glycoprotein HN in SDS-PAGE, corresponding to a decrease of about 4000 in its mol. wt. We propose that the PGJ2-induced alteration in the molecular structure of the HN protein prevents the insertion of this protein into the cell membrane thereby blocking virus maturation. The alpha,beta-unsaturated carbonyl group in the cyclopentane ring of the PGJ2 molecule may be necessary for antiviral activity.

Suppression of virus replication by prostaglandin A is associated with heat shock protein synthesis.
The antiviral action of cyclopentenone prostaglandins (PGs) is generally associated with alterations in the synthesis and/or maturation of specific virus proteins. In particular, inhibition of Sendai virus (SV) replication in African green monkey kidney cells by PGA1 has been shown to be a cell-mediated event, due to alterations in SV protein glycosylation and accompanied by the induction of a ...
متن کاملInhibition of poliovirus replication by prostaglandins A and J in human cells.
Cyclopentenone prostaglandins (PGs) inhibit the replication of a wide variety of enveloped DNA and RNA viruses. The antiviral activity is associated with alterations in the synthesis, maturation, and intracellular translocation of viral proteins. In the present report, we describe the effects of cyclopentenone PGs PGA1 and delta 12-PGJ2 on poliovirus (PV) replication in HeLa cells. Both PGs wer...
متن کاملInhibition of Foot-and-Mouth Disease Virus Replication by Hydro-alcoholic and Aqueous-Acetic Acid Extracts of Alhagi maurorum
Foot-and-mouth disease (FMD) is a major infectious disease of cloven-hoofed animals that is caused by the FMD virus (FMDV). This disease has significantly adverse economic impacts; therefore, rapid control measures are urgently. Traditional ranchers in Iran use Alhagi maurorum Medik. to treat FMD; therefore, we aimed to examine the antiviral activity of methanolic, ethanolic, and aqueous-acetic...

Objective: Influenza virus, which is associated with high level of morbidity and mortality, has been recently considered a public health concern; however, the methods of choice to control and treat it are limited. Our previous study showed anti-influenza virus activity of pomegranate peel extract (PPE). In this study, the mechanism through which PPE acts against influenza virus...
متن کاملProstaglandin A inhibits the replication of vesicular stomatitis virus: effect on virus glycoprotein.
Prostaglandins of the A series were found to strongly suppress the replication of vesicular stomatitis virus (VSV) in mouse L fibroblasts. The highest non-toxic dose of PGA1, 4 micrograms/ml, decreased VSV production by 93.6%. At this dose, PGA1 did not alter DNA, RNA or protein synthesis in uninfected L cells for periods up to 24 h, whereas it further suppressed protein synthesis and slightly ...
