Beta3-adrenoceptor activation just says NO to myocardial reperfusion injury.

چکیده
For the increasing number of patients who survive an acute myocardial infarction, infarct size is the main determinant of further prognosis (1). Timely reperfusion is mandatory to reduce infarct size, and for all conditioning strategies— ischemic pre-conditioning, delayed ischemic pre-conditioning, ischemic post-conditioning, and remote pre-conditioning— reperfusion is a prerequisite to reduce infarct size (i.e., they delay rather than ultimately prevent infarct development) (2). he pioneering experiments demonstrating reduction of inarct size by timely reperfusion are only 40 years old (3), and nfarct size reduction by a variety of experimental interventions mmediately became an attractive target then (4). However, the nitial enthusiasm for all sorts of interventions to reduce infarct ize soon gave way to a more sober and critical view (5). In ontrast, infarct size reduction by reperfusion through thromolysis initially and primary percutaneous intervention subseuently was quickly translated to clinical use in patients with cute myocardial infarction (2).

The negative inotropic effect of beta3-adrenoceptor stimulation is mediated by activation of a nitric oxide synthase pathway in human ventricle.
Beta1- and beta2-adrenoceptors in heart muscle cells mediate the catecholamine-induced increase in the force and frequency of cardiac contraction. Recently, in addition, we demonstrated the functional expression of beta3-adrenoceptors in the human heart. Their stimulation, in marked contrast with that of beta1- and beta2-adrenoceptors, induces a decrease in contractility through presently unkno...
متن کاملa-Adrenergic Mechanisms in Myocardial Ischemia
c-Adrenoceptor-mediated effects of sympathetic activation on the heart and coronary circulation are reviewed with emphasis on the pathophysiology of myocardial ischemia. A classification of a-adrenoceptor subtypes is presented, and the effects of a-adrenoceptor activation on presynaptic sympathetic nerve terminals, cardiomyocytes, endothelium, platelets, and coronary smooth muscle cells are dis...
متن کاملExploring the role of dimethylarginine dimethylaminohydrolase-mediated reduction in tissue asymmetrical dimethylarginine levels in cardio-protective mechanism of ischaemic postconditioning in rats
Objective(s): Reperfusion of ischaemic myocardium results in reduced nitric oxide (NO) biosynthesis by endothelial nitric oxide synthase (eNOS) leading to endothelial dysfunction and subsequent tissue damage. Impaired NO biosynthesis may be partly due to increased levels of asymmetrical dimethylarginine (ADMA), an endogenous inhibitor of eNOS. As dimethylarginine dimet...
متن کاملBeta3-adrenoreceptor stimulation ameliorates myocardial ischemia-reperfusion injury via endothelial nitric oxide synthase and neuronal nitric oxide synthase activation.
OBJECTIVES This paper examined whether nebivolol protects the heart via nitric oxide (NO) synthase and NO-dependent signaling in an in vivo model of acute myocardial infarction. BACKGROUND Beta(3)-adrenergic receptor (AR) activation promotes endothelial nitric oxide synthase (eNOS) activity and NO bioavailability. We hypothesized that specific beta(3)-AR agonists would attenuate myocardial is...
متن کاملThe effect of asafoetida essential oil on myocardial ischemic-reperfusion injury in isolated rat hearts
Objective: Previous studies reported that asafetida from Ferula assa-foetida Linn. species and its essential oil (AEO) have antioxidant effects. In the present study, the effect of AEO was evaluated on ischemic-reperfusion injury in isolated rat hearts. Materials and Methods: Forty-eight male Wistar rats were divided into 6 groups: 1) control group, 2) vehicle group, 3-5) AEO groups and, 6) car...
