چکیده

on a conventional ultramicrotome. Semi-thin sections on grids were stained with the Giemsa method and methylene blue-azure II-basic fuchsin. Staining without removal of the embedding matrix was sharp and brilliant. These stained sections were observed under a light microscope. For electron microscopy, they were doubly stained with uranyl acetate and lead salt. Identical sites on such sections could be distinctly examined with an accelerating potential of 100 kV at low magnification (250-1,500 times) using LEM-2000, a combined light and electron microscope. Thus, photomicrographs electron micrographs of identical sites on tissue samples could be compared exactly.

Light and electron microscopic observations of glycogen in semi-thin sections employing GMA, Quetol 523 and methylmethacrylate.

The use of improved GMA-Quetol 523 or GMA-Quetol 523 methyl methacrylate (MMA) mixture as an infiltration medium for the histochemical demonstration of glycogen has been devised to facilitate embedding, sectioning and staining. An improved method of infiltration uses such mixtures with a double weight of QCU-1 as a catalyst. Since the double weight of catalyst prevents interference by bleeding ...

A hydrophilic resin-embedding method for light and electron microscopic detection of tissue anionic sites with cationic colloidal iron: as applied to mouse Paneth cells.

A cationic colloidal iron method was introduced for electron microscopic detection of anionic sites in hydrophilic resin-embedded specimens, and the method was applied to Paneth cells of the mouse jejunum. Mouse jejunal blocks were embedded in hydrophilic acrylic resin (LR White), cut into ultrathin sections, stained with the diluted cationic colloidal iron, and exposed to osmium vapor. The jej...

A simple and reliable method for correlative light and electron microscopic studies.

We describe in detail a simple method for flat-embedding that can be subsequently used in correlative light and electron microscopic studies. The method can be applied to any material suitable for electron microscopy and is especially useful for study of the synaptology and ultrastructural characteristics of immunocytochemically or morphologically identified neurons or their processes. We prese...

Use of anti-fluorophore antibody to achieve high-sensitivity immunolocalizations of transporters and ion channels.

We have discovered that the immunoreactivity of the fluorophore Alexa Fluor 488 survives glutaraldehyde and osmium tetroxide fixation and epoxy resin embedding and etching. We have developed new localization methods that for the first time take advantage of this property. The antigen is localized in cryosections using suitable primary antibody and an Alexa Fluor 488-conjugated secondary antibod...

Oxidized p-phenylenediamine staining of epoxy resin sections.

Semithin and thin sections of glutaraldehyde-fixed, epoxy resin-embedded animal tissues were treated with solutions of oxidized p-phenylenediamine (PPD). This method is suitable to reveal the general morphology of tissues in light microscopy, showing a high staining degree in some polyanion containing components. Posttreatments of thin sections with gold chloride solutions give considerable ele...
