Near-Field Fluorescence Imaging and Fluorescence Lifetime Measurement of Light Harvesting Complexes in Intact Photosynthetic Membranes
نویسنده
چکیده
Fluorescence lifetime measurements with nanometer spatial resolution are made possible by incorporating time-correlated single photon counting with a near-field fluorescence microscope. Using this apparatus, single pieces of intact photosynthetic membrane from the Chlamydomonas reinhardtii strain doubly deficient in photosystem I (PS I) and photosystem I1 (PS 11) are examined. Comparison of simultaneous force and emission images shows single membrane bilayers lying flat on the mica substrate with the embedded light harvesting complex (LHC 11) proteins aggregated isotropicly across the membrane imaged. Fluorescence lifetimes are measured a t a single spot on the membrane with picosecond time resolution.

A Thin Layer Imaging with the Total Internal Reflection Fluorescence Microscopy
Total internal reflection fluorescence microscopy (TIRFM) is an optical technique that allows imaging of a thin layer of the sample with a thickness of about 100-200 nm. It is used in science of cell biology to study cellular processes, especially near the membranes of living cells. This method is based on the total internal reflection phenomenon, where the evanescent wave is generated in the l...
متن کاملStrong antenna-enhanced fluorescence of a single light-harvesting complex shows photon antibunching
The nature of the highly efficient energy transfer in photosynthetic light-harvesting complexes is a subject of intense research. Unfortunately, the low fluorescence efficiency and limited photostability hampers the study of individual light-harvesting complexes at ambient conditions. Here we demonstrate an over 500-fold fluorescence enhancement of light-harvesting complex 2 (LH2) at the single...
متن کاملMolecular architecture of plant thylakoids under physiological and light stress conditions: a study of lipid-light-harvesting complex II model membranes.
In this study, we analyzed multibilayer lipid-protein membranes composed of the photosynthetic light-harvesting complex II (LHCII; isolated from spinach [Spinacia oleracea]) and the plant lipids monogalcatosyldiacylglycerol and digalactosyldiacylglycerol. Two types of pigment-protein complexes were analyzed: those isolated from dark-adapted leaves (LHCII) and those from leaves preilluminated wi...
متن کاملPhotoprotective energy dissipation involves the reorganization of photosystem II light-harvesting complexes in the grana membranes of spinach chloroplasts.
Plants must regulate their use of absorbed light energy on a minute-by-minute basis to maximize the efficiency of photosynthesis and to protect photosystem II (PSII) reaction centers from photooxidative damage. The regulation of light harvesting involves the photoprotective dissipation of excess absorbed light energy in the light-harvesting antenna complexes (LHCs) as heat. Here, we report an i...

Time-resolved fluorescence spectroscopy was used to explore the pathway and kinetics of energy transfer in photosynthetic membrane vesicles (chromatophores) isolated from Rhodobacter (Rba.) sphaeroides cells harvested 2, 4, 6 or 24 hours after a transition from growth in high to low level illumination. As previously observed, this light intensity transition initiates the remodeling of the photo...
