چکیده

Objective: To study the expression levels of the NOK (novel oncogene with kinase-domain) gene in renal cell carcinoma (RCC) and its association with the progression of this cancer. Methods: In this study, immunohistochemistry (IHC) and Western blot analyses were applied to investigate the NOK expression level in RCC and adjacent normal renal tissue samples. MTT, colony formation, and migration assays were also utilized to evaluate the role of NOK in RCC cell lines. Results: Knocked-down expression of NOK in an RCC cell line (786-O) suppressed cellular proliferation and migration by restraining the activation of AKT and ERK. We found that the expression level of NOK was significantly higher in RCC tissues than in their adjacent tissues, and more importantly, overexpression of NOK was evidently correlated with the tumor TNM stage and Fuhrman grade (P<0.001). A high level of NOK was also associated with poor overall survival (P<0.05) and disease-free survival (P<0.05) by Kaplan-Meier analysis. Conclusions: NOK expression increased in RCC and was significantly correlated with TNM stage, Fuhrman grade, poor overall survival, poor disease-free survival, metastasis, and proliferation in RCC cells by regulating the activation of AKT and ERK, suggesting that NOK may play important roles as a positive regulator to RCC.

TRANSFORMING GROWTH FACTOR β2 UP-REGULATES GM-CSF GENE IN HUMAN BLADDER CARCINOMA CELL LINE HTB 5637

Transforming growth factor betas are multifunctional polypeptides in the cytokine superfamily. They have a growth inhibitory role on hemopoietic progenitor cells in semisolid colony assay as well as in long-term bone-marrow culture. TGF - β2 represses stromal cells, stem cell factor gene transcription, and decreases the stability of c-kit transcripts in hemopoietic cells. TGF-β also modulat...

Serine Arginine-Rich Splicing Factor 1 (SRSF1) Contributes to the Transcriptional Activation of CD3ζ in Human T Cells

T lymphocytes from many patients with systemic lupus erythematosus (SLE) express decreased levels of the T cell receptor (TCR)-associated CD3 zeta (ζ) signaling chain, a feature directly linked to their abnormal phenotype and function. Reduced mRNA expression partly due to defective alternative splicing, contributes to the reduced expression of CD3ζ chain. We previously identified by oligonucle...

Lactobacillus Acidophilus Cytotoxicity Effect and Apoptosis in Human Bladder Carcinoma Cells: An In Vitro Study

Introduction: Anticancer effects of Lactobacillus acidophilus, as a probiotic bacterium, have been indicated in several studies. There are common therapeutic options for bladder cancer treatment; however, their side effects and recurrence of disease are considerable. Therefore, complementary medication is essential and it must be safe and effective. In the present research, we assessed the anti...

Expression of Tra2β in Cancer Cells as a Potential Contributory Factor to Neoplasia and Metastasis

The splicing regulator proteins SRSF1 (also known as ASF/SF2) and SRSF3 (also known as SRP20) belong to the SR family of proteins and can be upregulated in cancer. The SRSF1 gene itself is amplified in some cancer cells, and cancer-associated changes in the expression of MYC also increase SRSF1 gene expression. Increased concentrations of SRSF1 protein promote prooncogenic splicing patterns of ...

The centrosomal kinase NEK2 is a novel splicing factor kinase involved in cell survival

NEK2 is a serine/threonine kinase that promotes centrosome splitting and ensures correct chromosome segregation during the G2/M phase of the cell cycle, through phosphorylation of specific substrates. Aberrant expression and activity of NEK2 in cancer cells lead to dysregulation of the centrosome cycle and aneuploidy. Thus, a tight regulation of NEK2 function is needed during cell cycle progres...
