چکیده

The spectroscopic responses to environmental changes (e.g. different solvents or temperatures) of the nitrile stretching transition of small molecule-nitriles or unstructured peptides were explored to better understand the observations of the nitrile-modified RNase complexes. In general, when solvated by a polar solvent, it is expected that the large ground state dipole moment of nitrile-containing molecules will exert an orienting force on the nearby solvent dipoles. These oriented dipoles will exert an electrostatic force on ground state dipole

Nitrile bonds as infrared probes of electrostatics in ribonuclease S.

Three different nitrile-containing amino acids, p-cyanophenylalanine, m-cyanophenylalanine, and S-cyanohomocysteine, have been introduced near the active site of the semisynthetic enzyme ribonuclease S (RNase S) to serve as probes of electrostatic fields. Vibrational Stark spectra, measured directly on the probe-modified proteins, confirm the predominance of the linear Stark tuning rate in desc...

Measuring electrostatic fields in both hydrogen-bonding and non-hydrogen-bonding environments using carbonyl vibrational probes.

Vibrational probes can provide a direct readout of the local electrostatic field in complex molecular environments, such as protein binding sites and enzyme active sites. This information provides an experimental method to explore the underlying physical causes of important biomolecular processes such as binding and catalysis. However, specific chemical interactions such as hydrogen bonds can h...

A solvatochromic model calibrates nitriles' vibrational frequencies to electrostatic fields.

Electrostatic interactions provide a primary connection between a protein's three-dimensional structure and its function. Infrared probes are useful because vibrational frequencies of certain chemical groups, such as nitriles, are linearly sensitive to local electrostatic field and can serve as a molecular electric field meter. IR spectroscopy has been used to study electrostatic changes or flu...

Ribonuclease S dynamics measured using a nitrile label with 2D IR vibrational echo spectroscopy.

A nitrile-labeled amino acid, p-cyanophenylalanine, is introduced near the active site of the semisynthetic enzyme ribonuclease S to serve as a probe of protein dynamics and fluctuations. Ribonuclease S is the limited proteolysis product of subtilisin acting on ribonuclease A, and consists of a small fragment including amino acids 1-20, the S-peptide, and a larger fragment including residues 21...

Electrostatic fields near the active site of human aldose reductase: 1. New inhibitors and vibrational stark effect measurements.

Vibrational Stark effect spectroscopy was used to measure electrostatic fields in the hydrophobic region of the active site of human aldose reductase (hALR2). A new hALR2 inhibitor was designed and synthesized that contains a nitrile probe with a Stark tuning rate of 0.77 cm-1/(MV/cm). Mutations to amino acid residues in the vicinity of the nitrile functional group were selected based on electr...
