Dardas, T. J. (Michigan State University, East Lansing), and V. H. Mallmann. Electrophoretic and immunoelectrophoretic studies of sera from normal, tuberculous, and noninfected tuberculin-sensitive guinea pigs. J. Bacteriol. 92:76-81. 1966.-Normal guinea pig serum was separated into seven fractions by electrophoresis on cellulose acetate membranes. Thirty antigens were found by immunoelectrophoresis: albumin, 6 alpha(1) globulins, 11 alpha(2) globulins, 6 beta(1) globulins, 5 beta(2) globulins, and gamma globulin. Hyper-alpha(2)-globulinemia was detected in sera from guinea pigs 14 days after inoculation with viable virulent Mycobacterium bovis. An additional alpha(2) globulin, not demonstrable prior to infection, was detected concomitantly with the hyper-alpha(2)-globulinemia by immunoelectrophoresis. The additional alpha(2) globulin was tentatively named alpha(2)-T. It persisted until the death of the guinea pigs. Neither hyper-alpha-globulinemia nor the alpha(2)-T antigen was detected by cellulose acetate electrophoresis and immunoelectrophoresis of sera from guinea pigs sensitized with heat-killed M. bovis. Both changes were due to the disease, not to delayed sensitivity alone.

The Effect of Tuberculin and Cortisone Singly and in Combination on Explanted Tissues of Guinea Pigs,
Previous experiments have shown that the growth of ceils explanted from tuberculin-sensitive animals may be inhibited if tuberculin is added to the culture menstrnum (1). Tissue culture techniques also have been employed in studies of the mechanism of action of cortisone. The direct effect of this compound upon such diverse physiologic phenomena as heart muscle contraction (2) and renal tubular...
متن کاملIsolation, characterization, and biological properties of a tuberculin-active peptidoglycan isolated from the culture filtrate of Mycobacterium tuberculosis.
A water-soluble tuberculin-active peptidoglycan (TAPG) with a molecular weight of ca. 28,000 to 30,000 was isolated from the culture filtrate of Mycobacterium tuberculosis. TAPG was approximately four to five times more potent than tuberculin purified protein derivative S in guinea pigs sensitized with M. tuberculosis or M. bovis (freeze-dried BCG). It showed little or no cross-reactivity at a ...
متن کاملTissue Culture Studies on Bacterial Allergy in Experimental Brucellosis
Tissue culture methods have been used to investigate infectious allergy in experimental brucellosis. A study was made of the effect of whole cell Brucella antigen on cultures of spleen from normal and Brucella-infected guinea pigs. The degree of toxicity was based upon the inhibition of migration of wandering cells and upon the morphologic appearance of stained sections of tissue cultures at di...
متن کاملHemorrhages in Tuberculous Guinea Pigs at the Site of Injection of Irritants following Intravascular Injections of Injurious Substances (shwartzman Phenomenon)
1. When toxic filtrates from cultures of B coli, B. typhosus, or meningococci are injected into the blood stream, peritoneal cavity, or subcutaneous tissue of tuberculous guinea pigs, the skin at the site of a tuberculin reaction becomes hemorrhagic. The extent of the hemorrhage is proportional to the severity of the tuberculin reaction demonstrable by tests with various dilutions of tuberculin...
متن کاملThe Effect of Tuberculin and Cortisone Singly and in Combination on Explanted Tissues of Guinea Pigs, Mice, and Rabbits
Tuberculin (P.P.D.) and cortisone acetate, singly and in combination, were added to explanted splenic tissue derived from guinea pigs, mice, and rabbits, and buffy coats obtained from rabbits. These tissues came from normal animals or from animals which had been infected with the tubercle bacillus or immunized with killed tubercle bacilli. 5 microg./ml. of tuberculin or 0.5 microg./ml. of corti...
