چکیده

Leupeptin~ a proteinase inhibitor obtained from culture f i l t rates of Streptomyces sp.~ has been proposed as a valuable agent for the restriction of proteolysis in vivo. We have previously shown tha t mammalian tissues possess an enzymic system that is capable of inactivating leupeptin (Beynon R39 Brown CP & Butler PEr 1981, Acta Biol. Med. Germ. @0, 153%1546). This paper demonstrates the ubiquitous distribution of the inactivating system throughout mammalian tissues and provides evidence for the location of the enzyme(s) in the soluble fraction of the cell.

Clonality of CD3 negative large granular lymphocyte proliferations determined by PCR based X-inactivation studies.

AIMS To examine persistent CD3-large granular lymphocytosis (LGL) cases for clonality, both by lineage specific (T cell receptor) and lineage independent (X-inactivation) molecular methods; and to find out whether X-inactivation studies are more appropriate than gene rearrangement studies for this subset of LGL disorders. METHODS Patients were selected who had LGL of more than six months' dur...

PTEN loss represses glioblastoma tumor initiating cell differentiation via inactivation of Lgl1

Glioblastoma multiforme is an aggressive and incurable type of brain tumor. A subset of undifferentiated glioblastoma cells, known as glioblastoma tumor initiating cells (GTICs), has an essential role in the malignancy of this disease and also appears to mediate resistance to radiation therapy and chemotherapy. GTICs retain the ability to differentiate into cells with reduced malignant potentia...

Involvement of the detoxifying enzyme lactoylglutathione lyase in Streptococcus mutans aciduricity.

Streptococcus mutans, a normal inhabitant of dental plaque, is considered a primary etiological agent of dental caries. Its main virulence factors are acidogenicity and aciduricity, the abilities to produce acid and to survive and grow at low pH, respectively. Metabolic processes are finely regulated following acid exposure in S. mutans. Proteome analysis of S. mutans demonstrated that lactoylg...

The Drosophila tumor suppressor gene lethal(2) giant larvae (lgl) encodes a cytoskeletal protein required for the change in shape and polarity acquisition of epithelial cells, and also for asymmetric division

Inactivation of the Drosophila tumor suppressor gene lethal(2)giant larvae (lgl; l(2)gl – FlyBase), leads to uncontrolled proliferation of the neuroblasts of the optic lobes and of the epithelial cells of the larval imaginal discs. The lgl mutant larvae never pupariate, and during the delayed larval stage disc cells become non-adhesive and undifferentiated, with no apparent polarity. When trans...

Nonselective autophagy of cytosolic enzymes by isolated rat hepatocytes

Seven cytosolic enzymes with varying half-lives (ornithine decarboxylase, 0.9 h; tyrosine aminotransferase, 3.1 h; tryptophan oxygenase, 3.3 h; serine dehydratase, 10.3 h; glucokinase, 12.7 h; lactate dehydrogenase, 17.0 h; aldolase, 17.4 h) were found to be autophagically sequestered at the same rate (3.5%/h) in isolated rat hepatocytes. Autophagy was measured as the accumulation of enzyme act...
