The Biosynthesis of Cytidine 5’-Monophospho-IV-acetylneuraminic Acid by an Enzyme from Neisseriu meni@i&s*

چکیده
In the accompanying paper (2) it was shown that Neisseria meningitidis, strain 1908 (Group C), forms N-acetylneuraminic acid by the condensation of N-acetyl-n-mannosamine and phosphoenolpyruvate with elimination of the phosphate group. In a preliminary communication we have reported a neK type of reaction carried out by another enzyme of Ncisserio meningitidis, strain 1998, which might be considered the activation of N-acetylneuraminic acid (1). In this irreversible reaction, cytidine triphosphate condenses with N-acetylneuraminic acid to form cytidine 5’-monophospho-N-acetylneuraminic acid (Diagram 1)’ and inorganic pyrophoaphate (Equation 1).

We have established an efficient method for enzymatic production of cytidine 5'-monophospho-N-acetylneuraminic acid (CMP-NeuAc) from inexpensive materials, N-acetylglucosamine (GlcNAc) and cytidine 5'-monophosphate (CMP). The accompanying paper (Heath and Ghalambor, 1963) presents evidence for the occurrence of 2-keto-3-deoxy-octonate @DO) as a glycosidically bound constituent in preparations of cell wall lipopolysaccharide (LPS) obtained from Escherichia cs 0111-B4. The purpose of this paper is to report the biosynthesis of cytidine monophospho-RDO (CMP-KDO) by an enzyme purified from this organism. Materials a...

Previous studies have reported that plants contain negligible amounts of free or protein-bound N-acetylneuraminic acid (Neu5Ac). This is a major disadvantage for the use of plants as a biopharmaceutical expression system, since N-glycans with terminal Neu5Ac residues are important for the biological activities and half-lives of recombinant therapeutic glycoproteins in humans. For the synthesis ...

It is known, human cells are incapable of synthesizing the N-glycolyl neuraminic acid (NeuGc) due to the inactivation of the cytidine monophospho-N-acetyl-neuraminic acid hydroxylase, the enzyme responsible for the synthesis of this sialic acid [1]. Conversely, the aberrant expression of NeuGc-sialoconjugates has been detected in humans, although preferentially in malignant tissues [2,3]. The m...
