2,4-dinitrophenyl Receptors on Mouse Thymus and Spleen Cells
نویسندگان
چکیده
A higher percentage of specific antigen-binding cells can be detected not only in normal CBA/J mouse spleen cell preparations (0.99%), but also in the normal thymus cell preparations (0.15%) with the use of [(125)I]2,4-dinitrophenyl-human IgG (DNP-HGG) as compared with most other antigens employed under similar conditions. The receptors on these cells are mainly specific for the DNP group as shown by the inhibition studies with DNP-lysine and the other DNP conjugates. In addition, it was shown by the inhibition studies with DNP-lysine that the thymus cells seem to have a lower avidity for DNP than the spleen cells. Preincubation of cell suspensions with antisera to immunoglobulins showed that the DNP-HGG antigen-binding cells in the thymus are inhibited predominantly with anti-micro-chain serum and the spleen cells with both anti-micro-chain and anti-gamma-chain sera; both cell populations were also significantly inhibited with the antisera to kappa-chains and Fab fragments. These data indicate that the nature of the receptor on the T cell differs from that on the majority of spleen cells.

Stimulation of Antibody Production to the Hapten 2,4-dinitrobenzene by Affinity Labeled Murine Lymphoid Cells
The ability of meta-nitrobenzenediazonium fluoborate (m-NBDF)-labeled thymus and spleen (S) cells to transfer immunity to 2,4-dinitrophenyl (DNP) into irradiated syngeneic recipients was investigated. There was a significant increase in the number of anti-DNP plaque-forming cells (PFC) when m-NBDF-labeled thymus cells and normal spleen cells, or normal thymus cells and m-NBDF-labeled spleen cel...
متن کاملActivation of T and B Lymphocytes in Vitro
In the presence of a preparation of purified thymus extract factors (TEF), spleen cells from athymic nude mice develop in vitro immune responses to soluble DNP-protein conjugates, to particulate sheep red blood cells, and to allogeneic cells. TEF has similar effects also on the in vitro antibody responses of normal mouse spleen cells, enhancing antigen-dependent plaque-forming cells responses t...
متن کاملCellular Requirements for the Primary in Vitro Antibody Response to Dnp-ficoll
The cellular requirements for the primary in vitro IgM and IgG response to dinitrophenyl-substituted Ficoll were examined. Neither thymus-derived lymphocytes nor macrophage-rich splenic adherent cells were required for anti-DNP antibody synthesis. DNP-Ficoll is therefore tentatively classified as a "thymic-independent" antigen.
متن کاملInduction of Immunological Tolerance to a Thymus-dependent Antigen in the Absence of Thymus-derived Cells
Specific immunological unresponsiveness was induced using thymus-dependent antigens in congenitally athymic (nu/nu) mice, in which no T-cell function has been demonstrated. The tolerance was induced in vivo by the injection of 5-10 mg of either FGG or DNP-HGG. Spleen cells from treated mice were tested in vitro for the ability to mount thymus-independent immune responses against FGG in the pres...
متن کاملIn Vitro Induction of a Primary Response to the Dinitrophenyl Determinant
Primary antibody response against the dinitrophenyl group has been elicited in vitro after the stimulation of normal mouse spleen explants with 2,4-dinitrophenyl (DNP)-hemocyanin or alpha-DNP-poly-L-lysine (PLL). Antibodies were detected in the culture medium by the inactivation of DNP-T4 phage. The specificity of the reaction was manifested by the lack of the capacity of the medium to inactiva...
