چکیده

Commensal Escherichia coli form biofilms at body temperature by expressing the extracellular matrix components curli fimbriae and cellulose. The role of curli fimbriae and cellulose in the interaction of commensal E. coli with the intestinal epithelial cell line HT-29 was investigated. Expression of curli fimbriae by the typical commensal isolate E. coli TOB1 caused adherence and internalization of the bacteria and triggered IL-8 production in HT-29 cells. In particular, induction of IL-8 production was complex and involved curli-bound flagellin. While cellulose alone had no effect on the interaction of TOB1 with HT-29 cells, co-expression of cellulose with curli fimbriae decreased adherence to, internalization and IL-8 induction of HT-29 cells. Investigation of a panel of commensal isolates showed a partial correlation between expression of curli fimbriae and enhanced internalisation and IL-8 production. Thus, the consequences of expression of extracellular matrix components on commensal bacterial-host interactions are complex.

Expression of cellulose and curli fimbriae by Escherichia coli isolated from the gastrointestinal tract.

Escherichia coli colonizes the gastrointestinal tract of humans; however, little is known about the features of commensal strains. This study investigated whether expression of the biofilm extracellular matrix components cellulose and curli fimbriae is found among commensal isolates. Fifty-two E. coli strains were isolated from faecal samples and, as a control, 24 strains from urinary tract inf...

Role of C-di-gmp Signalling in Bacterial-host Interactions

Bacteria have various ways to sense environmental signals and to adapt their behavior and physiology through different signaling systems. Secondary messenger signaling, amplified by enzymatic activity, rapidly transmits a signal in the cell resulting in allosteric functional control. Cyclic diguanosine monophosphate (c-di-GMP) is a novel global secondary messenger, that is found exclusively in ...

Anti-cancer properties of Escherichia coli Nissle 1917 against HT-29 colon cancer cells through regulation of Bax/Bcl-xL and AKT/PTEN signaling pathways

Objective(s): Chemotherapies used to treat colon cancer might often fail due to the emergence of chemoresistance and side effects. Escherichia coli Nissle 1917 (EcN) is a beneficial probiotic, whose molecular mechanisms in the prevention of colon cancer are yet to be fully understood. The present study assessed the anti-cancer effects of EcN treatments in human colorec...

The Ability of Cellulose Polysaccharide and Curli Pili Production in Uropathogenic Escherichia Coli and its Association with Biofilm Formation Intensity

Abstract        Background and Objective: the Formation of urinary infection by uropathogenic E.coli needs   numerous virulence factors and biofilm formation is among these factors. Bacteria that form biofilms express phenotype traits that appear according to the bacteria type. Cellulose is an important compound on the outside of E.coli causing bacte...

Alterations of c‐di‐GMP turnover proteins modulate semi‐constitutive rdar biofilm formation in commensal and uropathogenic Escherichia coli

Agar plate-based biofilm of enterobacteria like Escherichia coli is characterized by expression of the extracellular matrix components amyloid curli and cellulose exopolysaccharide, which can be visually enhanced upon addition of the dye Congo Red, resulting in a red, dry, and rough (rdar) colony morphology. Expression of the rdar morphotype depends on the transcriptional regulator CsgD and occ...
