Development of a tropoelastin-binding MR contrast agent for in vivo imaging of impaired elastogenesis in atherosclerosis

چکیده
Background Elastin is a primary component of the vessel wall and present in all elastic vessels such as the aorta and the pulmonary artery. Elastogenesis begins with the synthesis and secretion of the soluble precursor tropoelastin that becomes cross-linked into insoluble elastin. Under normal conditions cross-linked elastin is the only form of the molecule present in the vessel wall whereas tropoelastin is absent. Conversely, under pathological conditions including atherosclerosis and aortic aneurysms elastogenesis resumes and tropoelastin molecules accumulate in the vessel wall. We developed a novel tropoelastin-binding MRI contrast-agent that would allow 1) specifically bind to tropoelastin but not to cross-linked mature elastin to allow 2) detection of pathologic elastogenesis that occurs in atherosclerosis and 3) minimizing non-specific signal from endogenously present mature elastin.

Recent Advances in PET-MR Hybrid contrast agent
Introduction: All of the Imaging modalities have advantages and disadvantages alone. So if we want to have the best and perfect image, combining these modalities produces something we desired. PET-MR images consist of morphologic and metabolic data. MRI and PET provide high spatial and contrast resolution and high sensitivity and molecular information respectively. Hybrid PET-...
متن کاملAssessment of Myocardial Remodeling Using an Elastin/Tropoelastin Specific Agent with High Field Magnetic Resonance Imaging (MRI)
BACKGROUND Well-defined inflammation, proliferation, and maturation phases orchestrate the remodeling of the injured myocardium after myocardial infarction (MI) by controlling the formation of new extracellular matrix. The extracellular matrix consists mainly of collagen but also fractions of elastin. It is thought that elastin is responsible for maintaining elastic properties of the myocardium...

Objective: Measurement of blood-brain permeability dysfunction in active and chronic MS lesions with T1-weighted dynamic contrast-enhanced MRI to show variation in inflammatory activity Background: blood-brain-barrier perfusion characterization impaired in MS as some studies have shown recently buta comparison between perfusion parameters in contrast-enhanced and non-enhanced lesions not have ...
متن کاملDynamic Contrast Magnetic Resonance Imaging (DCE-MRI) and Diffusion Weighted MR Imaging (DWI) for Differentiation between Benign and Malignant Salivary Gland Tumors
Background: Salivary gland tumors form nearly 3% of head and neck tumors. Due to their large histological variety and vicinity to facial nerves, pre-operative diagnosis and differentiation of benign and malignant parotid tumors are a major challenge for radiologists. Objective: The majority of these tumors are benign; however, sometimes they tend to transform into a malignant form. Functional M...
متن کاملSynthesis, Radiolabeling, and Biological Evaluation of Peptide LIKKPF Functionalized with HYNIC as Apoptosis Imaging Agent
Introduction: A noninvasive method of detecting exposure of phosphatidylserine (PS) on the external surface of the plasma membrane such as nuclear imaging could assist the diagnosis and therapy of apoptosis related pathologies. The most studied imaging agent for apoptosis is Annexin V so far. Because of limitations of Annexin V other agents have been introduced such as small peptides and molecu...
