Comparison of human colonic mucoprotein antigen from normal and neoplastic mucosa.

چکیده
Colonie mucoprotein antigen (CMA), a high-molecularweight, tissue-specific mucin, was obtained from 9 normal mucosa! samples and 12 colonie adenocarcinomas. Puri fication was achieved by means of phenokwater extrac tion followed by ethanol precipitation and molecular sieve chromatography. Chemical analyses of the individual nor mal CMA preparations indicated similar qualitative and quantitative compositions. The peptide portion of the molecule was characterized by a high content of threonine, serine, proline, glutamic acid, and glycine. The oligosaccharide chains were composed of W-glycolylneuraminic acid, fucose, galactose, glucosamine, and galactosamine. A comparison of the normal and tumor CMA revealed the following differences. Although the amino acid compositions were similar, tumor CMA's had 40% less threonine and 40% more aspartic acid than did CMA's from normal tissue. Both observations were statistically significant at 2 p < 0.001. Normal and tumoral mucins could be distinguished by the aspartate:threonine ratio with a confidence of greater than 99.9%. A 2-fold increase in molar ratio of sialic acid:threonine in tumor-derived CMA was detected (2p 0.05). These differences suggest that tumor CMA has a lower density of carbohydrate moieties on the peptide core. Tumor CMA may arise by the expression of a new or altered DNA sequence. Alter natively, the possibility exists that the normal colonie mucosa synthesizes more than one mucin and that, as a consequence of neoplasia, one (or more) or these is suppressed or enhanced.

Immunoperoxidase localization of colonic mucoprotein antigen in neoplastic tissues.
The distribution of colonic mucoprotein antigen in various types of cancer was examined to determine whether the antigen might have a role as a tissue marker. An antiserum prepared against normal colon-derived colonic mucoprotein antigen and appropriately absorbed with mucinous ovarian cystadenocarcinoma fluid had reactivity limited to the colon and cecum when tested by immunoperoxidase against...

متن کاملExpression of cloned sequences in biopsies of human colonic tissue and in colonic carcinoma cells induced to differentiate in vitro.
A computer-based scanning and image-processing system has been developed to quantitate the relative level of expression of each of 4000 cloned complementary DNA sequences in small biopsies routinely removed from the mucosa of normal and neoplastic human large intestine. Individuals have been studied from well-defined population groups in which colonic epithelial cells have progressed to increas...
متن کاملExpression of HLA class I antigens in sporadic adenomas and histologically normal mucosa of the colon.
The loss of HLA antigens by neoplastic cells is considered important for tumor growth and metastasis, inasmuch as it may allow tumors to escape immune surveillance. We have observed reduced expression of HLA antigens in sporadic colon cancer and adenomas from familial adenomatous polyposis patients. We now studied the expression of HLA class I antigens in patients with sporadic adenomas, which ...
متن کاملExpression of c-myb protooncogene and other cell cycle-related genes in normal and neoplastic human colonic mucosa.
The expression of c-myb, c-myc, histone H3, and ornithine decarboxylase genes was examined by Northern blot analysis in the normal and neoplastic mucosa of ten subjects affected by colon cancer. The mRNA levels of c-myb protooncogene were detected at low levels in all normal samples but were increased in the neoplastic mucosa of six cases in comparison to the normal counterpart. In five of thes...
