چکیده

Spores of Bacillus subtilis are being used as probiotics and competitive exclusion agents for animal consumption. Commercial production media often include relatively expensive components of animal origin that are a potential source for the presence of adventious agents, therefore undesirable for use in production scale. In this study a new animal-free component, chemically defined medium, was tested for B. subtilis spore production. Medium composition was optimized with respect to vitamin composition, carbon, nitrogen and calcium concentrations. A fed-batch bioprocess was developed, being the effect on sporulation of the carbon to nitrogen ratio at the end of the exponential growth phase studied. The developed strategy consisted of an initial and a final batch phase and an intermediate fed-batch phase with the addition of a feeding solution containing glucose and calcium and the addition of a feeding solution of ammonium sulphate, using an exponential and a constant feeding profile, respectively. Using the fed-batch strategy, it was possible to achieve a maximum spore production of 3.6 × 1010 spores/mL, corresponding to a 5 folds increase when compared to the preliminary batch experiments.

Single, chemically defined sporulation medium for Bacillus subtilis: growth, sporulation, and extracellular protease production.

The composition and application of a single, chemically defined medium or growth and sporulation of Bacillus subtilis is described. At 37 degrees C cells grew with a doubling time of about 40 min; cultures attained near-maximal spore formation (70 to 80% by 12 h after the end of exponential growth and produced 1 X 10(9) to 2 X 10(9) heat-resistant free spores at 24 h. Dipicolinic acid productio...

Extracellular control of spore formation in Bacillus subtilis.

Spore formation in the Gram-positive bacterium Bacillus subtilis has been classically viewed as an example of unicellular differentiation that occurs in response to nutritional starvation. We present evidence that B. subtilis produces an extracellular factor(s) that is required, in addition to starvation conditions, for efficient sporulation. This factor is secreted and accumulates in a cell de...

Single cell analysis of gene expression patterns of competence development and initiation of sporulation in Bacillus subtilis grown on chemically defined media.

AIM Understanding the basis for the heterogeneous (or bistable) expression patterns of competence development and sporulation in Bacillus subtilis. METHODS AND RESULTS Using flow cytometric analyses of various promoter-GFP fusions, we have determined the single-cell gene expression patterns of competence development and initiation of sporulation in a chemically defined medium (CDM) and in bio...

A new chemically defined medium for the growth and sporulation of Bacillus cereus strains in anaerobiosis.

A new chemically defined liquid medium, MODS, was developed for the aerobic growth and anaerobic growth and sporulation of Bacillus cereus strains. The comparison of sporulation capacity of 18 strains of B. cereus has shown effective growth and spore production in anaerobiosis..
