چکیده

plastic epidermis and in lesions of squamous cell carcinoma. The growth fraction in control unirradiated mice was also 100%. The duration of TV and S decreased progressively with induction and progression of the tumor. For normalappearing epidermis of irradiated mice and epidermis of control mice, TVand S were approximately 95 and 6.2 hr, respectively. With the induction of hyperplasia, TV and S were reduced to 43.75 and 4.45 hr, respectively, while even lower values were obtained for squamous cell carcinoma where TV = 16 hr and S = 3.05 hr. The labeling index in normal epidermis was 6.75% and increased progressively in hyperplastic epidermis (10.17%) and squamous cell carci noma (18.98%). These results indicate that ultraviolet light-induced epidermal tumor production in hairless mice is associated with a progressive shortening of the cell cycle.

Ultraviolet Light B-Mediated Inhibition of Skin Catalase Activity Promotes Gr-1+CD11b+ Myeloid Cell Expansion

Skin cancer incidence and mortality are higher in men compared with women, but the causes of this sex discrepancy remain largely unknown. UV light exposure induces cutaneous inflammation and neutralizes cutaneous antioxidants. Gr-1(+)CD11b(+) myeloid cells are heterogeneous bone marrow-derived cells that promote inflammation-associated carcinogenesis. Reduced activity of catalase, an antioxidan...

Silibinin prevents ultraviolet radiation-caused skin damages in SKH-1 hairless mice via a decrease in thymine dimer positive cells and an up-regulation of p53-p21/Cip1 in epidermis.

Non-melanoma skin cancer (NMSC) accounts for >1 million new cases each year in the US alone suggesting that more approaches are needed for its prevention and control. Earlier studies by us have shown that silymarin (a crude form of biologically active silibinin with some other isomers), isolated from milk thistle, affords strong protection against ultraviolet (UV) radiation-induced NMSC in SKH-...

Reduction of UV-induced skin tumors in hairless mice by selective COX-2 inhibition.

UV light is a complete carcinogen, inducing both basal and squamous cell skin cancers. The work described uses the selective COX-2 inhibitor celecoxib to examine the efficacy of COX-2 inhibition in the reduction of UV light-induced skin tumor formation in hairless mice. UVA-340 sun lamps were chosen as a light source that effectively mimics the solar UVA and UVB spectrum. Hairless mice were irr...

Oncogenes and Tumor Suppressors MIF Antagonist (CPSI-1306) Protects against UVB-Induced Squamous Cell Carcinoma

Macrophage migration inhibitory factor (MIF) is a homotrimeric proinflammatory cytokine implicated in chronic inflammatory diseases and malignancies, including cutaneous squamous cell carcinomas (SCC). To determinewhetherMIF inhibition could reduceUVB light–induced inflammation and squamous carcinogenesis, a small-molecule MIF inhibitor (CPSI-1306) was utilized that disrupts homotrimerization. ...

Effects of chronic low-dose ultraviolet B radiation on DNA damage and repair in mouse skin.

Chronic exposure to sunlight causes skin cancer in humans, yet little is known about how habitual exposure to low doses of ultraviolet B radiation (UVB) affects DNA damage in the skin. We treated Skh-1 hairless mice with daily doses of suberythemal UVB for 40 days and analyzed the amount and distribution of DNA photodamage using RIAs and immunofluorescence micrography. We found that DNA damage ...
