چکیده

• In influenza-immune subjects, H1N1 influenza virus–containing immune complexes can activate platelets through FcgRIIA. • H1N1 can also activate platelets through thrombin, independently of FcgRIIA, pointing to a role of coagulation in influenza infection. Platelets play crucial functions in hemostasis and the prevention of bleeding. During H1N1 influenza A virus infection, platelets display activation markers. The platelet activation

PLATELETS AND THROMBOPOIESIS Regulator of G-protein signaling 18 integrates activating and inhibitory signaling in platelets

Regulator of G-protein signaling 18 (RGS18) is a GTPase-activating protein for the G-q and G-i subunits of heterotrimeric Gproteins that turns off signaling by G-protein coupled receptors. RGS18 is highly expressed in platelets. In the present study, we show that the 14-3-3 protein binds to phosphorylated serines 49 and 218 of RGS18. Platelet activation by thrombin, thromboxaneA2, orADP stimula...

Regular Article PLATELETS AND THROMBOPOIESIS Bacillus anthracis peptidoglycan activates human platelets through FcgRII and complement

In humans, sepsis is caused by an exaggerated inflammatory response to components of infectious organisms. The inflammation involves platelets which propagate blood coagulation in the vasculature. Thrombocytopenia resulting from in vivo platelet aggregation is common in patients in intensive care units with bacterially driven sepsis and is prognostic for intensive care unit mortality. Thrombocy...

In vivo effects of lipopolysaccharide and TLR4 on platelet production and activity: implications for thrombotic risk.

Gram-negative bacteria release LPS, which activates Toll-like-receptor-4 (TLR4) in the host, initiating an inflammatory response to infection. Infection increases risk for thrombosis. Platelets contribute to defense from infection and to thrombosis. Experiments were designed to determine whether LPS, through TLR4 signaling, affects platelet phenotype. Platelet responses in wild-type (WT) mice a...

Regular Article PLATELETS AND THROMBOPOIESIS Q:A1 Myosin-II repression favors pre/proplatelets but shear activation generates platelets and fails in macrothrombocytopenia

• Myosin-II inhibition (with blebbistatin) and MYH9-RD mutations enhance shear fragmentation to pre/ proplatelet sizes. • Sustained shear activates normal myosin-II, which then favors division of pre/ proplatelets to smaller platelets. Megakaryocyte ploidy and the generation of pre/proplatelets are both increased in culture by pharmacologic inhibition of myosin-II, but nonmuscle myosin-IIA (MII...

Regular Article PLATELETS AND THROMBOPOIESIS Both TMEM16F-dependent and TMEM16F-independent pathways contribute to phosphatidylserine exposure in platelet apoptosis and platelet activation

Transbilayer lipid asymmetry in platelets and other cells is maintained by an aminophospholipid translocase, an active transporter pumping aminophospholipids phosphatidylserine (PS) and phosphatidylethanolamine from the outer to the inner leaflet of the plasma membrane. Under particular conditions, phospholipid asymmetry is disrupted by a “scrambling” process, in which the phospholipids become ...
