Reductive dechlorination of tetrachloroethene by a mixed bacterial culture growing on ethyl lactate.

چکیده
Chloroethenes like tetrachloroethene (PCE) are the most prevalent groundwater contaminants in the USA. Their presence as nonaqueous phase liquids (NAPLs) makes remediation difficult. Among options for NAPL cleanup, co-solvent injection has demonstrated success. However, the process has the potential to leave considerable residue of the co-solvent as well as residual chloroethene. Our rationale in this study was to examine whether this residual solvent could be a potential electron donor for the remediation of the residual chloroethene. We hypothesized that ethyl lactate, a "green" solvent, could serve both as a NAPL extraction solvent and an electron donor for reductive dechlorination of residual chloroethene. We examined whether a mixed culture known to degrade PCE with lactate could also grow on ethyl lactate and whether it could stimulate PCE dechlorination. Biomass growth and PCE dechlorination were observed by protein and chloride production, respectively, in the culture; with a specific dechlorination rate of 50 150 microg (mg cell d)(-1). Ethyl lactate abiotically breaks down to ethanol and lactate, the latter being a rich source of hydrogen fo reductive dechlorination. The results demonstrate that ethyl lactate may be promising for in situ bioremediation following NAPL extraction.

Identification of Multiple Dehalogenase Genes Involved in Tetrachloroethene-to-Ethene Dechlorination in a Dehalococcoides-Dominated Enrichment Culture
Chloroethenes (CEs) are widespread groundwater toxicants that are reductively dechlorinated to nontoxic ethene (ETH) by members of Dehalococcoides. This study established a Dehalococcoides-dominated enrichment culture (designated "YN3") that dechlorinates tetrachloroethene (PCE) to ETH with high dechlorination activity, that is, complete dechlorination of 800 μM PCE to ETH within 14 days in the...
متن کاملIdentification of a reductive tetrachloroethene dehalogenase in Shewanella sediminis.
The genome sequence of psychrophilic Shewanella sediminis revealed the presence of five putative reductive dehalogenases (Rdhs). We found that cell extracts of pyruvate/fumarate-grown S. sediminis cells catalysed reduced methyl viologen-dependent reductive dechlorination of tetrachloroethene (PCE) to trichloroethene (TCE) at a specific activity of approximately 1 nmol TCE min(-1) (mg protein)(-...
متن کاملReductive dechlorination of high concentrations of tetrachloroethene to ethene by an anaerobic enrichment culture in the absence of methanogenesis.
Tetrachloroethene, also known as perchloroethylene (PCE), is a common groundwater contaminant throughout the United States. The incomplete reductive dechlorination of PCE--resulting in accumulations of trichloroethene, dichloroethene isomers, and/or vinyl chloride--has been observed by many investigators in a wide variety of methanogenic environments. Previous mixed-culture studies have demonst...
متن کاملHydrogen as an electron donor for dechlorination of tetrachloroethene by an anaerobic mixed culture.
Hydrogen served as an electron donor in the reductive dechlorination of tetrachloroethene to vinyl chloride and ethene over periods of 14 to 40 days in anaerobic enrichment cultures; however, sustained dechlorination for more extended periods required the addition of filtered supernatant from a methanol-fed culture. This result suggests a nutritional dependency of hydrogen-utilizing dechlorinat...
متن کاملA New Electron Donor for Reductive Dechlorination of Perchloroethylene
One of the main problems associated with remediation of chlorinated solvent groundwater contamination is the presence of NAPLs in the source zone. NAPLs are often better treated through solvent extraction, surfactant washing, or vapor extraction than through bioremediation. Removal of NAPL through solvent extraction would likely leave a substantial residual of both the extraction solvent and th...
