Construction of a genetic linkage map in the wild plant Mimulus using RAPD and isozyme markers.

چکیده
As a first step to mapping quantitative trait loci for mating system differences, a genetic linkage map was generated from an interspecific backcross between Mimulus guttatus and Mimulus platycalyx. The linkage map consists of 99 RAPD and two isozyme markers. Eighty-one of these markers were mapped to 15 linkage groups, spanning 1437 contiguous centiMorgans, and covering 58% of the estimated genome. The genome length of Mimulus is estimated at 2474 +/- 35 cM; bootstrapping indicates that only ca. 40 markers are needed to give an accurate estimate of genome length. Further statistical analyses indicate that many RAPD markers cannot be ordered with certainty and that uncertain linkage groups tend to map nonlinearly even under commonly used mapping functions. Strategies for speeding up the mapping process for a wild species and possible applications of a partial linkage map in evolutionary studies are discussed. Key words : linkage map, mating system, Mimulus, RAPD.

Linkage Map Construction for Silkworm (Bombyx mori L.) Based on Amplified Fragment Length Polymorphism Markers
The domesticated silkworm, Bombyx mori, is of high commercial importance as a silk producer and is also widely used for implementation of basic and applied research. It is important to understand its genome organization using molecular markers for genetic studies and for breeding purposes. In this study, a genetic linkage map using 204 amplified fragment length polymorphism (AFLP) markers was d...

The combined use of the arbitrarily primed polymerase chain reaction [AP-PCR, also known as random amplification of polymorphic DNA (RAPD}] and isozyme mapping resulted in the production of 87 potential marker loci, enabling an overall expansion within the genetic map of the fish genus Xiphophorus. Use of DNA sequencing-style acrylamide gels and carefully controlled conditions of amplification ...

The wild mint "Mentha longifolia (L.) Hudson subsp. noeana (Briq.) Briq." is an important medicinal plant which contains several worthy components like: menthone, pulegone, pipertenon, menthol, thymol, and carvone in its essential oil. This plant grows extensively in center of Asia including Iran. Because of so worthy components and its big role in traditional and modern medicine, the wild mint...
متن کاملIdentification of QTLs for grain yield and some agro-morphological traits in sunflower (Helianthus annuus L.) using SSR and SNP markers
Many agriculturally important traits are complex, affected by many genes and the environment. Quantitative trait loci (QTL) mapping is a key tool for studying the genetic structure of complex traits in plants. In the present study QTLs associated with yield and agronomical traits such as leaf number, leaf length, leaf width, plant height, stem and head diameter were identified by using 70 recom...

A detailed genetic map has been constructed from the F2 population of a single intraspecific cross of Lactuca sativa (n = 9). It comprises 319 loci, including 152 restriction fragment length polymorphism (RFLP), 130 random amplified polymorphic DNA (RAPD), 7 isozyme, 19 disease resistance, and 11 morphological markers. Thirteen major, four minor linkage groups and several unlinked markers are i...
