Molecular mechanisms behind TRIM 28 expression

TRIM28 is an interferon regulated gene, possibly playing a part in regulating the IFN system involved in the defence against pathogens. However, when type I IFNs are overexpressed it can lead to autoimmune diseases. We investigate how TRIM28 expression is regulated on transcriptional and post-transcriptional levels by exploring transcription factors involved in regulating transcription when bound to the promoter region of TRIM28 and by studying miRNAs targeting TRIM28 transcripts. We also investigated direct effects of IFN-β on proposed regulatory agents in vivo. Our results show that the transcription factor E2F1 and miR-582 are strong candidates for TRIM28 expression regulation and that IFN-β do not regulate E2F1 or miR-582 in vivo.
