The Kinetics of Coupled Enzyme Reactions

نویسنده

چکیده

1. A theory is developed to account for the kinetics of coupled-enzyme reactions without assuming that the second reaction follows first-order kinetics. 2. A simple procedure is described for applying the theory to the practical design ofenzyme assays. 3. The validity of the theory is confirmed for the assay of glucokinase with glucose 6-phosphate dehydrogenase as coupling enzyme. 4. The possibility of extending the theory to three or more coupled reactions is discussed.

The Kinetics of Enzyme Mixtures

Even purified enzyme preparations are often heterogeneous. For example, preparations of aspartate aminotransferase or cytochrome oxidase can consist of several different forms of the enzyme.  For this reason we consider how different the kinetics of the reactions catalysed by a mixture of forms of an enzyme must be to provide some indication of the characteristics of the species present.  Based...

Multi-Dimensional Simulation of n-Heptane Combustion under HCCI Engine Condition Using Detailed Chemical Kinetics

In this study, an in-house multi-dimensional code has been developed which simulates the combustion of n-heptane in a Homogeneous Charge Compression Ignition (HCCI) engine. It couples the flow field computations with detailed chemical kinetic scheme which involves the multi-reactions equations. A chemical kinetic scheme solver has been developed and coupled for solving the chemical reactions an...

Coupled Enzyme Reactions Performed in Heterogeneous Reaction Media: Experiments and Modeling for Glucose Oxidase and Horseradish Peroxidase in a PEG/Citrate Aqueous Two-Phase System

The intracellular environment in which biological reactions occur is crowded with macromolecules and subdivided into microenvironments that differ in both physical properties and chemical composition. The work described here combines experimental and computational model systems to help understand the consequences of this heterogeneous reaction media on the outcome of coupled enzyme reactions. O...

Determining Enzyme Kinetics for Systems Biology with Nuclear Magnetic Resonance Spectroscopy

Enzyme kinetics for systems biology should ideally yield information about the enzyme's activity under in vivo conditions, including such reaction features as substrate cooperativity, reversibility and allostery, and be applicable to enzymatic reactions with multiple substrates. A large body of enzyme-kinetic data in the literature is based on the uni-substrate Michaelis-Menten equation, which ...

Multistep Surface Electrode Mechanism Coupled with Preceding Chemical Reaction-Theoretical Analysis in Square-Wave Voltammetry

In this theoretical work, we present for the first time voltammetric results of a surface multistep electron transfer mechanism that is associated with a preceding chemical reaction that is linked to the first electron transfer step. The mathematical model of this so-called “surface CEE mechanism” is solved under conditions of square-wave voltammetry. We present relevant set of results portrayi...
