Vary, J. C. (University of Wisconsin, Madison), and H. O. Halvorson. Kinetics or germination of Bacillus spores. J. Bacteriol. 89:1340-1347. 1965.-The kinetics of germination of Bacillus cereus strain T spores was accurately described by McCormick. To study the mechanism of germination, it is necessary to correlate the characteristic changes in a population of germinating spores with the behavior of the individual spores in the same population. Two microscopic events are apparent during germination: microlag, the time interval between the addition of l-alanine to heat-activated spores and the beginning of loss in refractility, and microgermination time, the time for the actual change in refractility to occur. The frequency distributions of both events are skewed, and appear to be independent. The effects of l-alanine concentration, heat activation, and temperature of germination on three parameters, microlag, microgermination, and per cent germination, were microscopically studied. The data are discussed in relation to the mechanism of germination, and a correlation between microlag and microgermination times with the constants of McCormick's equation has been suggested.

Kinetics of Germination of Individual Spores of Geobacillus stearothermophilus as Measured by Raman Spectroscopy and Differential Interference Contrast Microscopy
Geobacillus stearothermophilus is a gram-positive, thermophilic bacterium, spores of which are very heat resistant. Raman spectroscopy and differential interference contrast microscopy were used to monitor the kinetics of germination of individual spores of G. stearothermophilus at different temperatures, and major conclusions from this work were as follows. 1) The CaDPA level of individual G. ...

Keynan, A. (University of Wisconsin, Madison) and H. O. Halvorson. Calcium dipicolinic acid-induced germination of Bacillus cereus spores. J. Bacteriol. 83:100-105. 1962.-The germination of spores of Bacillus cereus strain T can be initiated by calcium dipicolinic acid. The kinetics of germination are characterized by a long lag period followed by a rapid loss of refractility. The lag period di...
متن کاملMonitoring the kinetics of uptake of a nucleic acid dye during the germination of single spores of Bacillus species.
Dormant bacterial spores do not take up and bind nucleic acid dyes in the spore core but readily take up such dyes when they are fully germinated. We present a methodology that combines fluorescence microscopy, phase contrast microscopy, and laser tweezers Raman spectroscopy to monitor the kinetics of uptake of the nucleic acid dye SYTO 16 during germination of individual Bacillus cereus and Ba...
متن کاملInvolvement of the spore coat in germination of Bacillus cereus T spores.
Bacillus cereus T spores were prepared on fortified nutrient agar, and the spore coat and outer membrane were extracted by 0.5% sodium dodecyl sulfate-100 mM dithiothreitol in 0.1 M sodium chloride (SDS-DTT) at pH 10.5 (coat-defective spores). Coat-defective spores in L-alanine plus adenosine germinated slowly and to a lesser extent than spores not treated with SDS-DTT, as determined by decreas...
متن کاملMonitoring rates and heterogeneity of high-pressure germination of bacillus spores by phase-contrast microscopy of individual spores.
Germination of Bacillus spores with a high pressure (HP) of ∼150 MPa is via activation of spores' germinant receptors (GRs). The HP germination of multiple individual Bacillus subtilis spores in a diamond anvil cell (DAC) was monitored with phase-contrast microscopy. Major conclusions were that (i) >95% of wild-type spores germinated in 40 min in a DAC at ∼150 MPa and 37°C but individual spores...
