Monoxenic Culture of Banana-Parasitic Nematodes on Musa acuminata cv. Poyo shoots.

چکیده
Among nematodes parasitizing bananas throughout the world, Radopholus similis and Helicotylenchus multicinctus are the most widespread and damaging species. Besides these two nematodes, Hoplolaimus pararobustus is encountered frequently in Ivory Coast and has been increasing in banana growing areas for the past 25 years (6,10). In vitro culture systems could facilitate screening for resistance and rearing of large numbers of monoxenic nematodes for experiments. The most common techniques used to rear R. similis monoxenically involve callus tissues, usually carrot (13). Root callus from okra, grapefruit, and alfalfa (12) and leaf callus from citrus (9) also have been utilized. On the other hand, the burrowing nematode has been mass produced on differentiated tissues such as excised okra roots (7). Radopholus similis was cultured on citrus roots produced from leaves (9), on citrus seedlings growing in sandy soil irrigated with nutrient solution (5), and on citrus roots growing in an agar medium (8). Brown and Vessey (2) demonstrated that R. similis thrives on banana fruit callus. Helicotylenchus multicinctus, however, failed to survive on the same callus. There is no

Aggressiveness and Damage Potential of Central American and Caribbean Populations of Radopholus spp. in Banana.
Monoxenic cultures of burrowing nematode populations extracted from banana roots from Belize, Guatemala, Honduras, and Costa Rica were established on carrot discs. Cultures of Radopholus spp. were also obtained from Florida, Puerto Rico, Dominican Republic, and Ivory Coast. The aggressiveness (defined as reproductive fitness and root necrosis) of these populations was evaluated by inoculating b...
متن کاملAnalysis of Induced Mutants of Salinity Resistant Banana (Musa acuminata cv. Dwarf Cavendish) Using Morphological and Molecular Markers
Genetic diversity amongst 21 induced mutant clones tolerant to salinity along with one non-irradiated sensitive clone of banana (Musa acuminata cv. Dwarf Cavendish (AAA)) were studied using morphological and random amplified polymorphic DNA (RAPD) markers. Out of the 30 phenotypic indices screened, 23 were polymorph and two traits, leaf habit and blotches color, were differentiated by non-irrad...
متن کاملEffect of benzylaminopurine (BAP) pulsing on in vitro shoot multiplication of Musa acuminata (banana) cv. Berangan
MS (Murashige and Skoog) media supplemented with benzylaminopurine (BAP) showed that the number of bud formation in shoot cultures of Musa acuminata cv.Berangan during the initiation stage increased proportionately with the concentrations used (11, 22 and 33 μM). However, the highest concentration of BAP (33 μM) simultaneously increased the formation of abnormal shoots. After the first apical b...

Phenylphenalenone-type compounds accumulated in the tissues of two banana cultivars-Musa acuminata cv. "Grande Naine" (AAA) and Musa acuminata × balbisiana Colla cv. "Bluggoe" (ABB)-when these were fed on by the banana weevil (Cosmopolites sordidus (Germ.) (Coleoptera: Curculionidae)) and the banana stem weevil (Odoiporus longicollis (Oliver) (Coleoptera: Curculionidae)). The chemical constitue...
متن کاملEffects of Liquid Medium on Rooting and Acclimation of Regenerated Microshoots of Banana (Musa sapientum L.) cv. Sagar
A regeneration protocol ensuring a high frequency rooting of micro-shoots derived from apical and axillary buds of suckers of banana cv. Sagar was achieved by using liquid medium. When the explants were cultured on MS medium supplemented with 0.5 mg/l each of BA, Kn and NAA, a large number of shoots developed. With the progression of the number of subcultures, shoot proliferation was enhanced. ...
