چکیده

Download ral noncoding RNAs do vital cellular functions, including gene regulation and cell differentiation. Pre, we reported that vault RNA (vRNA) has the ability to recognize chemotherapeutic compounds, such oxantrone, based on biophysical and biochemical analyses. In the present study, we show that human stoma-, leukemia-, and osteocarcinoma-derived cell lines overexpress vRNA and exhibit higher resistoward mitoxantrone. Interestingly, when vRNA expression was suppressed by RNA interference in cells, the resistance progressively decreased. In agreement with these findings, overexpression of -1 caused resistance to mitoxantrone. These results suggest a role of vRNA in mitoxantrone resistance in malignant cells and justify further studies on the importance and application of noncoding RNAs in cancer chemotherapeutics. Expression of noncoding vault RNA in human malignant cells and its importance in mitoxantrone resistance.

Several noncoding RNAs do vital cellular functions, including gene regulation and cell differentiation. Previously, we reported that vault RNA (vRNA) has the ability to recognize chemotherapeutic compounds, such as mitoxantrone, based on biophysical and biochemical analyses. In the present study, we show that human glioblastoma-, leukemia-, and osteocarcinoma-derived cell lines overexpress vRNA...

Linkage between Large intergenic non-coding RNA regulator of reprogramming and Stemness State in Samples with Helicobacter pylori Infection of Gastric Cancer Cells

Background: Long noncoding RNAs (lncRNAs), as non-protein coding transcripts, play key roles in tumor progression and stemness state in many malignancies, as their aberrant expression has been found in gastric cancer (GC) as one of the most common cancer worldwide. LINC-ROR (large intergenic noncoding RNA regulator of reprogramming) identified as an involved lncRNA in human malignancies, howeve...

NSun2-Mediated Cytosine-5 Methylation of Vault Noncoding RNA Determines Its Processing into Regulatory Small RNAs

Autosomal-recessive loss of the NSUN2 gene has been identified as a causative link to intellectual disability disorders in humans. NSun2 is an RNA methyltransferase modifying cytosine-5 in transfer RNAs (tRNAs), yet the identification of cytosine methylation in other RNA species has been hampered by the lack of sensitive and reliable molecular techniques. Here, we describe miCLIP as an addition...

HOTAIR Induces the Downregulation of miR-200 Family Members in Gastric Cancer Cell Lines

Background: Gastric cancer (GC) is the fourth most common human malignancy and the second reason for cancer morbidity worldwide. Long noncoding RNA (LncRNA) HOX transcript antisense RNA (HOTAIR) has recently emerged as a promoter of metastasis in various cancer types, including GC, through the epithelial‑mesenchymal transition (EMT) process. However, the exact mechanism of HOTAIR in promoting E...

Study of Long Noncoding RNA FER1L4 and RB1, as Its Competing Endogenous RNA Network Target Gene, in Breast Cancer

Introduction: Breast cancer is the second most common cause of cancer-related death among females, which requires an exploration for markers to propose a more specific categorization of this cancer. Long noncoding RNAs (lncRNAs), the main subset of noncoding transcripts, are involved in tumorigenic processes. In this study, we investigated the expression of the fer-1–like family member 4 (FER...
