چکیده

Received for publication January 22, 1965. were fixed after injection were determined by the use of †̃25I-labeledantibody and radioautography and were found to be mainly in connective tissues and occasionally on sinusoid walls (5). The inicrosome fraction of cells is derived from the endoplasmic reticulum, which has been claimed to be contiguous to cell surface. Moreover, Wallach and Hager have shown in the case of Erhlich ascites cells that this fraction contains antigens associated with cell surface (8). Since the localization of antibodies on cells would occur most effectively on cell surface, studies were under taken to examine antibodies against the microsome fraction of rat hepatoma. Our previous report clearly indicated that antibodies against hepatoma microsomes did localize in the hepatomas and in normal livers when injected (4).

Differences between the microsomes of normal rat liver and of N-2-fluorenylacetamide-induced rat hepatoma as determined by the paired label antibody technic.

Rabbit antibodies against the microsome fractions of a chemi cally induced rat hepatoma and of normal rat liver were com pared by the paired label radioantibody technic. The antihopatoma antibodies were labeled with 125Iand the anti-normal-liver antibodies were labeled with 131I.The 2 were mixed and then in jected or treated in vitro simultaneously. The 2 antibodies differed in vivo in their lo...

Purification of localizing antibodies formed against microsomes of the N-2-fluorenylacetamide-induced rat hepatoma.

ment of the N-2-fluorenylacetamide-induced rat hepa tomas have been shown to localize in livers with hepatoma as well as in normal livers when injected i.v. (3—5, 7). The localization was shown by the use of radioiodine and fluo resceili labels. Since localization of antibodies requires direct contact of the antibody with the structure on which it is fixed, we investigated the properties of a...

Increased activity of polynucleotide ligase from rat hepatoma induced by N-2-fluorenylacetamide.

Polynucleotide ligase in both the soluble fraction and the nuclear extract, which catalyzes the covalent joining of 2 segments of an interrupted strand in a DNA duplex, was studied with normal rat liver and hepatoma induced by yV-2-fluorenylacetamide. The activity of this enzyme in hepatoma was about 5 times greater in the soluble fraction and about 3 times greater in the nuclear fraction than ...

MICROSOME-MEDIATED BENZO[A]PYRENE-DNA BINDING AND INHIBITION BY CYTOSOLIC FRACTIONS FROM LIVER AND SKIN OF ADULT AND WEANLING RATS

Biotransformation of benzo[a]pyrene (BaP) in the presence of microsomal fractions derived from liver and epiderm of adult and weanling rats was examined. The aim of this study was to evaluate the effect of age on the capacity of two organs in transformation of BaP. Subcellular fractions were prepared from skin and liver by ultracentrifugation and were used as the source of BaP metabolizing enzy...

Relative in vitro stability of ornithine decarboxylase from liver preneoplastic nodules and hepatomas.

A very rapid and drastic microsome-dependent in vitro inactivation of the hydrocortisone-induced ornithine decarboxylase in rat liver was reported recently (M. F. Zuretti and E. Gravela, Biochim. Biophys. Acta. 742: 269-277, 1983). Present results show that ornithine decarboxylase from preneoplastic nodules and hepatomas, which have been induced in rats by N-2-fluorenylacetamide, is much more s...
