Cardiac Alterations in Mice with Altered GLUT4 Expression
نویسندگان
چکیده
Genetic ablation of GLUT4, the major insulin sensitive transporter, results in insulin resistance without overt diabetes. GLUT4 Null mice exhibit significantly decreased adipose tissue and severe cardiac hypertrophy (2x Control) while maintaining normal whole body glycemia. The hypertrophy seen in Null animals is not due to pressure overload. Histological examination revealed enlarged myocytes and the presence of ischemic damage in Null cardiac tissue. GLUT4 Null hearts also suffer from metabolic abnormalities, which could potentiate the hypertrophy. MRI and Langendorff perkion studies of the Null and Control hearts were performed to determine the degree of hypertrophy and hemodynamic performance under normal and ischemic conditions using substrates containing glucose.

Preservation of glucose metabolism in hypertrophic GLUT4-null hearts.
GLUT4-null mice lacking the insulin-sensitive glucose transporter are not diabetic but do exhibit abnormalities in glucose and lipid metabolism. The most striking morphological consequence of ablating GLUT4 is cardiac hypertrophy. GLUT4-null hearts display characteristics of hypertrophy caused by hypertension. However, GLUT4-null mice have normal blood pressure and maintain a normal cardiac con...
متن کاملFailure of insulin-regulated recruitment of the glucose transporter GLUT4 in cardiac muscle of obese Zucker rats is associated with alterations of small-molecular-mass GTP-binding proteins.
Cardiac ventricular tissue of lean and genetically obese (fa/fa) Zucker rats was used to study the expression, subcellular distribution and insulin-induced recruitment of the glucose transporter GLUT4 and to elucidate possible molecular alterations of the translocation process. Hearts were removed from basal and insulin-treated (20 min) lean and obese Zucker rats, and processed for subcellular ...
متن کاملReduced Slc2a4/GLUT4 expression in subcutaneous adipose tissue of monosodium glutamate obese mice is recovered after atorvastatin treatment
BACKGROUND Decreased expression of glucose transporter protein GLUT4, encoded by the solute carrier 2A4 (Slc2a4) gene, is involved in obesity-induced insulin resistance. Local tissue inflammation, by nuclear factor-κB (NFκB)-mediated pathway, has been related to Slc2a4 repression; a mechanism that could be modulated by statins. Using a model of obesity with insulin resistance, this study invest...
متن کاملCardiac hypertrophy with preserved contractile function after selective deletion of GLUT4 from the heart.
Glucose enters the heart via GLUT1 and GLUT4 glucose transporters. GLUT4-deficient mice develop striking cardiac hypertrophy and die prematurely. Whether their cardiac changes are caused primarily by GLUT4 deficiency in cardiomyocytes or by metabolic changes resulting from the absence of GLUT4 in skeletal muscle and adipose tissue is unclear. To determine the role of GLUT4 in the heart we used ...
متن کاملAnkyrin-B metabolic syndrome combines age-dependent adiposity with pancreatic β cell insufficiency.
Rare functional variants of ankyrin-B have been implicated in human disease, including hereditary cardiac arrhythmia and type 2 diabetes (T2D). Here, we developed murine models to evaluate the metabolic consequences of these alterations in vivo. Specifically, we generated knockin mice that express either the human ankyrin-B variant R1788W, which is present in 0.3% of North Americans of mixed Eu...
