چکیده

It is known from the work of Elwyn et al. (1) that ethanolamine is incorporated as a unit into the choline molecule during the synthesis de novo of this substance. Du Vigneaud et al. (2, 3) showed that at least some of the methyl groups of choline are derived directly from the methyl group of methionine; and in an earlier communication we provided evidence that all the methyl groups originate from this source (4). Recently, as a result of studies with rat liver in viva (5) and in vitro (6, 7), Bremer and Greenberg proposed that choline is formed by the stepwise transfer of three methyl groups from X-adenosyl methionine to phosphotidyl ethanolamine. The work reported here essentially confirms their findings and offers some further support for the reactions which they formulated.

Effects of analogles of ethanolamine and choline on phospholipid metabolism in rat hepatocytes.

1. Analogues of ethanolamine and choline were incubated with different labelled precursors of phospholipids and isolated hepatocytes and the effects on phospholipid synthesis were studied. 2. 2-Aminopropan-1-ol and 2-aminobutan-1-ol were the most efficient inhibitors of [(14)C]ethanolamine incorporation into phospholipids, whereas the incorporation of [(3)H]choline was inhibited most extensivel...

Effect of cytidine on membrane phospholipid synthesis in rat striatal slices.

Using rat striatal slices, we examined the effect of cytidine on the conversion of [3H]choline to [3H]-phosphatidylcholine ([3H]PC), and on net syntheses of PC, phosphatidylethanolamine (PE), and phosphatidylserine, when media did or did not also contain choline, ethanolamine, or serine. Incubation of striatal slices with cytidine (50-500 microM) caused dose-dependent increases in intracellular...

Regulation of Phospholipid Metabolism in Differentiating Cells from Rat Brain Cerebral Hemispheres in Culture

Cultured dissociated cells from rat embryo cerebral hemisphere iqcorporate [gH]and [U-14C]ethanolamine into cellular lipids. Nearly all radioactivity in the lipid fractions is incorporated into 1,2diacylethanolamine phosphoglycerides and I-alkenyl,P-acylethanolamine phosphoglycerides (plasmalogen). Kinetic data suggest that the rate of labeling of both ethanolamine phospholipids from the phosph...

Sterol carrier protein-2 expression alters phospholipid content and fatty acyl composition in L-cell fibroblasts.

The effects sterol carrier protein-2 (SCP-2) expression on L-cell phospholipid levels and fatty acyl composition was assessed using L-cells transfected with the murine cDNA encoding for either the 15 kDa proSCP-2 or 13.2 kDa SCP-2. Expression of these proteins reduced total phospholipid mass (nmol/mg protein) by 24% and reduced the cholesterol to phospholipid ratio 60 and 28%, respectively. In ...
