چکیده

A viral agent inducing syncytia formation in monolayer cul tures of bovine embryonic spleen cells and a rabbit cornea cell line was isolated from cattle with lymphosarcoma. The agent was strongly cell-associated and transmissible with cell-free ff1trates at very low dilutions. An antigen prepared from heavily infected cultures produced precipitin lines by immunodiffu sion when tested against sera from lymphosarcomatous and apparently normal cattle. Buffy coat cells and cellular dc ments present in milk from apparently normal cattle whose sera gave positive precipitin reactions contained the virus. Some nonreacting offspring of dams that reacted were also virus carriers. Fluorescent conjugates prepared with precipita ring sera stained infected cells specifically while nonprecipita ting sera failed to do so. Electron microscopic examination of ultrathin sections from infected cell cultures revealed virus-like particles budding at the cell membranes in a manner resem bling the avian and murine leukemia viruses. Morphologically, these virus-like particles bore some resemblance to the mouse mammary tumor virus. Evidence that the virus plays a role, if any, in inducing bovine lymphosarcoma has not been estab lished.

Induction of syncytia by the bovine C-type leukemia virus.

Bovine buffy coat cells infected with the bovine leukemia virus (BLV) induce syncytia formation in human diploid embryonic lung cells as well as in monolayer cell cultures of bovine, simian, ovine, bat, and caprine origin, but not in mouse fibroblast cells, normall rat kidney cells, or RSV-transformed rat cells. Syncytia were not observed in diploid embryonic lung cells inoculated with bovine b...

Dual effect of interleukin 4 on HIV-1 expression: implications for viral phenotypic switch and disease progression.

We report that interleukin 4 (IL-4) inhibits the propagation of non-syncytia-inducing and increases the propagation of syncytia-inducing HIV-1 isolates by two mechanisms. It differentially regulates the two major HIV-1 coreceptors, CCR5 and CXCR4, in human peripheral blood mononuclear cells, increasing CXCR4 and decreasing CCR5 expression in primary CD4(+) T-lymphocytes. In addition, IL-4 stimu...

P-87: Effects of Bovine Cumulus Cell Co-Cul tire on Post Fertilization Development of Buffalo (Bubalus bubalis) Oocyte

Background: There is a considerable numbers of buffalo (Bubalus bubalis) in Iran which have an important role in rural life. The reproductive performance of buffalo is lower than cow. Co-culture with somatice cells improved the maturation rate of most of the species. The aim of the present study was to evaluate the effect of bovine cumulus co-culture on post fertilization development of buffalo...

Syncytia induction enhances the oncolytic potential of vesicular stomatitis virus in virotherapy for cancer.

Vesicular stomatitis virus (VSV) selectively replicates in tumor but not in normal cells and is being developed as an oncolytic agent for cancer therapy. Here we report the construction of a recombinant VSV capable of inducing syncytia formation between tumor cells through membrane fusion at neutral pH, which led to enhanced oncolytic properties against multifocal hepatocellular carcinoma (HCC)...

Studies on human KC cell syncytia formation induced by Mason-Pfizer monkey virus.

Human KC cell monolayer inoculated with concentraten Mason-Pfizer monkey virus (MPMV) showed syncytia formation within an hour. The cell fusion was blocked by the treatment of the MPMV with neutralizing antiserum. Treatment of the MPVM with beta-propiolactone resulted in the loss of infectivity although KC cell fusion ability of the virus still remained. KC cells inoculated with unconcentrated ...
