چکیده

This work presents the design, fabrication, and testing of a cross-flow filtration microdevice, for the continuous extraction of bacteria from a whole blood sample for the treatment of sepsis and bacteremia. The system consists of a two-compartment microdevice with two aligned sets of PDMS microchannels, separated by a porous polycarbonate (PCTE) membrane with 2 m pores. When a whole blood sample flows through the channels on one side of the membrane (Reservoir channels), the larger blood cells are sterically excluded from passing through the membrane while the smaller bacteria are removed by traversing the membrane into the filtration channels.

Elasto-inertial microfluidics for bacteria separation from whole blood for sepsis diagnostics

BACKGROUND Bloodstream infections (BSI) remain a major challenge with high mortality rate, with an incidence that is increasing worldwide. Early treatment with appropriate therapy can reduce BSI-related morbidity and mortality. However, despite recent progress in molecular based assays, complex sample preparation steps have become critical roadblock for a greater expansion of molecular assays. ...

A microfluidics approach towards high-throughput pathogen removal from blood using margination.

Sepsis is an adverse systemic inflammatory response caused by microbial infection in blood. This paper reports a simple microfluidic approach for intrinsic, non-specific removal of both microbes and inflammatory cellular components (platelets and leukocytes) from whole blood, inspired by the invivo phenomenon of leukocyte margination. As blood flows through a narrow microchannel (20 × 20 µm), d...

Continuous-flow microfluidic blood cell sorting for unprocessed whole blood using surface-micromachined microfiltration membranes.

White blood cells (WBCs) constitute about 0.1% of the blood cells, yet they play a critical role in innate and adaptive immune responses against pathogenic infections, allergic conditions, and malignancies and thus contain rich information about the immune status of the body. Rapid isolation of WBCs directly from whole blood is a prerequisite for any integrated immunoassay platform designed for...

Dielectrophoretic separation of platelets from diluted whole blood in microfluidic channels.

The dielectrophoresis (DEP) phenomenon is used to separate platelets directly from diluted whole blood in microfluidic channels. By exploiting the fact that platelets are the smallest cell type in blood, we utilize the DEP-activated cell sorter (DACS) device to perform size-based fractionation of blood samples and continuously enrich the platelets in a label-free manner. Cytometry analysis reve...
