چکیده

PGE 2 exerts potent diuretic and natriuretic effects on the kidney. This action is mediated in part by direct inhibition of collecting duct Na 1 absorption via a Ca 11 -coupled mechanism. These studies examine the role the Ca 11 -coupled PGE-E EP 1 receptor plays in mediating these effects of PGE 2 on Na 1 transport. Rabbit EP 1 receptor cDNA was amplified from rabbit kidney RNA. Nuclease protection assays demonstrated highest expression of EP 1 mRNA in kidney, followed by stomach, adrenal, and ileum. In situ hybridization, demonstrated renal expression of EP 1 mRNA was exclusively over the collecting duct. In fura-2–loaded microperfused rabbit cortical collecting duct, EP 1 active PGE analogs were 10–1,000-fold more potent in raising intracellular Ca 11 than EP 2 , EP 3 , or EP 4 -selective compounds. Two different EP 1 antagonists, AH6809 and SC19220, completely blocked the PGE 2 -stimulated intracellular calcium increase. AH6809 also completely blocked the inhibitory effect of PGE 2 on Na 1 absorption in microperfused rabbit cortical collecting ducts. These studies suggest that EP 1 receptor activation mediates PGE 2 -dependent inhibition of Na 1 absorption in the collecting duct, thereby contributing to its natriuretic effects. ( J. Clin. Invest . 1998. 102:194–201.)

Prostaglandin E receptors and the kidney.

Prostaglandin E(2) is a major renal cyclooxygenase metabolite of arachidonate and interacts with four G protein-coupled E-prostanoid receptors designated EP(1), EP(2), EP(3), and EP(4). Through these receptors, PGE(2) modulates renal hemodynamics and salt and water excretion. The intrarenal distribution and function of EP receptors have been partially characterized, and each receptor has a dist...

Prostaglandin E2 in the Regulation of Water Transport in Renal Collecting Ducts

The kidney plays a central role in the regulation of the body water balance. The process of targeting the water channel aquaporin-2 (AQP2) on the apical plasma membrane of the collecting duct (CD) principal cells is mainly regulated by the antidiuretic peptide hormone arginine vasopressin (AVP), which is responsible for the maintenance of water homeostasis. Recently, much attention has been foc...

Prostaglandin E-prostanoid4 receptor mediates angiotensin II-induced (pro)renin receptor expression in the rat renal medulla.

Angiotensin II (Ang II) stimulates (pro)renin receptor (PRR) expression in the renal collecting duct, triggering the local renin response in the distal nephron. Our recent study provided evidence for involvement of cyclooxygenase-2-prostaglandin E2 pathway in Ang II-dependent stimulation of PRR expression in the collecting duct. Here, we tested the role of E-prostanoid (EP) subtypes acting down...

Urine concentrating defect in prostaglandin EP1-deficient mice.

We investigated the role of the prostaglandin E(2) (PGE(2)) EP(1) receptor in modulating urine concentration as it is expressed along the renal collecting duct where arginine-vasopressin (AVP) exerts its anti-diuretic activity, and in the paraventricular and supraoptic nuclei of the hypothalamus where AVP is synthesized. The urine osmolality of EP(1)-null mice (EP(1)(-/-)) failed to match level...

Epidermal growth factor inhibits amiloride-sensitive sodium absorption in renal collecting duct cells.

The effects of the ERK pathway on electrogenic transepithelial Na(+) absorption by renal collecting duct cells were determined. Approximately 90% of the unstimulated short-circuit current (15 +/- 1 microA/cm(2), n = 10) across conditionally immortalized murine collecting duct epithelial cells (mCT1) is amiloride sensitive and is likely mediated by apical epithelial Na(+) channels. Chronic expos...
