Weiss, Emilio (Naval Medical Research Institute, Bethesda, Md.) and Harry R. Dressler. Increased resistance to chloramphenicol in Rickettsia prowazekii with a note on failure to demonstrate genetic interaction among strains. J. Bacteriol. 83:409-414. 1962.-A further effort to produce genetically labeled Rickettsia prowazekii was made by serially passing the Madrid E strain in eggs in the presence of increasing concentrations of chloramphenicol. After 40 serial and 1 limit-dilution passages, a substrain, called C(r), was isolated, which appeared to be the result of a two-step change, each involving a twofold increase in resistance to the antibiotic. Chloramphenicol resistance was retained after 10 drugless egg passages. Cross-resistance to thiocymetin, but not to unrelated drugs, was shown. Several attempts were made to demonstrate genetic interaction, in eggs or entodermal cell cultures, among strains resistant to chloramphenicol, erythromycin, and p-aminobenzoic acid. They have not been successful.

Protein and RNA synthesis by isolated Rickettsia prowazekii.

Purified host-cell-free Rickettsia prowazekii synthesized protein from leucine, lysine, or proline as radioactive precursor. Most, if not all, of the rickettsial proteins were synthesized. The specific activities of the newly synthesized proteins in the cytoplasmic fraction and in the envelope fraction were not significantly different, indicating that both soluble and envelope proteins were syn...

Transformation of Rickettsia prowazekii to rifampin resistance.

Rickettsia prowazekii, the causative agent of epidemic typhus, is an obligate intracellular parasitic bacterium that grows directly within the cytoplasm of the eucaryotic host cell. The absence of techniques for genetic manipulation hampers the study of this organism's unique biology and pathogenic mechanisms. To establish the feasibility of genetic manipulation in this organism, we identified ...

Establishment of a Replicating Plasmid in Rickettsia prowazekii

Rickettsia prowazekii, the causative agent of epidemic typhus, grows only within the cytosol of eukaryotic host cells. This obligate intracellular lifestyle has restricted the genetic analysis of this pathogen and critical tools, such as replicating plasmid vectors, have not been developed for this species. Although replicating plasmids have not been reported in R. prowazekii, the existence of ...

Expression of Rickettsia Adr2 protein in E. coli is sufficient to promote resistance to complement-mediated killing, but not adherence to mammalian cells

Bacteria exposed to host serum are subject to the antibacterial effects to the complement system. However, pathogenic microorganisms have evolved mechanisms of evading this immune attack. We have previously demonstrated that at least two R. conorii antigens, RC1281/Adr1 and OmpB β-peptide, contribute to the evasion of complement-mediated killing by binding the complement regulatory proteins vit...

Effect of mouse lymphokines and cloned mouse interferon-gamma on the interaction of Rickettsia prowazekii with mouse macrophage-like RAW264.7 cells.

We studied the effects of crude mouse lymphokines and cloned mouse interferon-gamma on the interaction of Rickettsia prowazekii with mouse macrophage-like RAW264.7 cells. Treatment of RAW264.7 cells with lymphokines before infection, after infection, or both before and after infection with R. prowazekii led to killing of a substantial proportion of the RAW264.7 cells. Such cytotoxicity required...
