Cyclophosphamide-induced chimerism.

چکیده
BALB/c mice were injected with cyclophosphamide (Cytoxan) (180-280 mg/kg) i.p., and with 3 X 10T (BALB/c 9 X DBA a ) Fj (CDF) spleen cells i.v. The recipients were bled periodically and their sera tested for DBA y-globulin. All mice had levels of donor y-globulin at 2 weeks; 17/24 still had detectable levels at 6 months. A greater proportion of mice had detectable donor y-globulin for a longer time and in higher titer when treated with the highest dose of Cytoxan. All mice received CDF skin grafts 2 weeks after Cytoxan and CDF spleen infusion. Marked prolongation of skin graft survival was observed. In general, skin graft rejection correlated well with loss of donor y-globulin. However, 4 mice which rejected skin grafts within one month continued to have detectable donor y-globulin for 6 months. The results are similar to those re ported for X-irradiation-induced chimeras.

Posttransplant administration of cyclophosphamide and donor lymphocyte infusion induces potent antitumor immunity to solid tumor.
PURPOSE Nonmyeloablative allogeneic stem cell transplantation (SCT) has been increasingly used for the treatment of hematologic and solid malignancies, and mature donor T cells are considered to be the main effectors of the graft-versus-tumor (GVT) activity. However, the association between degree of donor chimerism and intensity of GVT effects has not been fully elucidated. We recently propose...
متن کاملRenal cancer treatment with low levels of mixed chimerism induced by nonmyeloablative regimen using cyclophosphamide in mice.
Recently, much attention has been paid to nonmyeloablative allogeneic stem cell transplantation for the treatment of metastatic renal cancer. Mature donor T cells cause graft-versus-host disease (GVHD) although they are also the main mediators of the beneficial graft-versus-tumor activity associated with this treatment. Hence, the segregation of the graft-versus-tumor activity from GVHD is an i...
متن کاملInduction of permanent mixed chimerism and skin allograft tolerance across fully MHC-mismatched barriers by the additional myelosuppressive treatments in mice primed with allogeneic spleen cells followed by cyclophosphamide.
A pure method of drug (cyclophosphamide plus busulfan)-induced skin allograft tolerance in mice that can regularly overcome fully H-2-mismatched barriers in mice has been established. The components of the method are i.v. administration of 1 x 108 allogeneic spleen cells on day 0, i.p. injection of 200 mg/kg CP and 25 mg/kg busulfan on day 2, and i.v. injection of T cell-depleted 1 x 107 bone m...
متن کاملRegulatory roles of NKT cells in the induction and maintenance of cyclophosphamide-induced tolerance.
We have previously reported the sequential mechanisms of cyclophosphamide (CP)-induced tolerance. Permanent acceptance of donor skin graft is readily induced in the MHC-matched and minor Ag-mismatched recipients after treatment with donor spleen cells and CP. In the present study, we have elucidated the roles of NKT cells in CP-induced skin allograft tolerance. BALB/c AnNCrj (H-2(d), Lyt-1.2, a...
متن کاملToward minimal conditioning protocols for allogeneic chimerism in tolerance resistant recipients
Mixed chimerism is a promising approach toward generating donor-specific immunological tolerance. However, chimerism induction can be toxic; therefore, there is an effort to develop non-myeloablative, minimal intensity protocols that can generate chimerism without the toxic side effects. Recently, with the goal of creating a minimalistic chimerism induction protocol in the tolerance resistant n...
متن کاملInduction of early post-transplant graft-versus-leukemia effects using intentionally mismatched donor lymphocytes and elimination of alloantigen-primed donor lymphocytes for prevention of graft-versus-host disease.
Graft-versus-leukemia (GVL) effects can be induced in tolerant mixed chimeras prepared with nonmyeloablative conditioning. GVL effects can be amplified by post-grafting donor lymphocyte infusion (DLI). Unfortunately, DLI is frequently associated with graft-versus-host disease (GVHD). We investigated the feasibility of induction of potent GVL effects by DLI using intentionally mismatched lymphoc...
