چکیده

INTRODUCTION Clopidogrel bisulfate, chemically S (+)-2-(2chlorophenyl)-6, 7-dihydrothiene [3,2-C]pyridine-5(4H)acetic acid methyl ester sulphate is a potent antiplatelets and antithrombotic drug (at C-7 clopidogrel has an absolute S configuration). Clopidogrel has been approved and marketed globally with the primary indication being to reduce atherosclerotic events in patients with several comorbid conditions due to stroke, myocardial infarction and cardiovascular disease. Clopidogrel displays a selective binding on the platelet surface to adenylate cyclase-coupled ADP receptors, which is unique as compared with aspirin and other related drugs. The mechanism of action of aspirin or other related agents involves inhibition of arachidonic acid metabolism, inhibition of cyclo oxygenase enzyme system or the inhibition of prostaglandin synthesis. Reports have suggested that clopidogrel does not have a direct effect on the enzyme systems cAMP phosphodiesterase, thromboxane A2, adenylate cyclase, as well no effects on the synthesis of prostacyclin [1].

Influence of methadone on clopidogrel in addicts on methadone maintenance therapy

Background: Clopidogrel is a prodrug that converts in the liver to an active thiol metabolite, which irreversibly inhibits the platelet P2Y12 adenosine diphosphate receptor. It seems that methadone as CYP2C19 inhibitor affects ticlopidine activity in vivo. This study aimed to test the ability of methadone in changing ticlopidine pharmacokinetics. Methods: We conducted a case–control st...

Development of Microemulsion for Solubility Enhancement of Clopidogrel

Clopidogrel, an inhibitor of platelet aggregation, selectively inhibits the binding of adenosine diphosphate (ADP) to its platelet receptor and the subsequent ADP-mediated activation of the glycoprotein GPIIb/IIIa complex, thereby inhibiting platelet aggregation. Oral bioavailability of clopidogrel is very low (less than 50%), due to its poor water solubility. The aim of this investigation was ...

Association of ABCB1 Gene Polymorphisms and Clopidogrel Responsiveness in Iranian Patients undergoing Percutaneous Coronary Intervention

Clopidogrel is an antiplatelet agent currently used for preventing stent thrombosis. Despite certain clinical benefits of clopidogrel in patients undergoing percutaneous coronary intervention (PCI), adequate antiplatelet effect has not been obtained in some patients. The present study was designed to investigate the potential association of ABCB1 (ATP-Binding Cassette, Subfamily B, member1) gen...

The Effect of Enoxaparin and Clopidogrel on Survival of Random Skin Flap in Rat Animal Model

BACKGROUND Necrosis of skin flaps is considered as an important complication in reconstructive surgery. We conducted an experimental study to investigate the efficacy of low-molecular weight heparin, clopidogrel and their combination to improve the flap survival. METHODS Forty male, adult Sprague-Dawlay rats were divided randomly into 4 groups. Standard rectangular, distally based dorsal ran...

Clopidogrel in non-ST segment elevation acute coronary syndromes: an overview of the submission by the British Cardiac Society and the Royal College of Physicians of London to the National Institute for Clinical Excellence, and beyond.

A comprehensive appraisal was undertaken on behalf of the British Cardiac Society and the Royal College of Physicians of London to assess the use of clopidogrel in acute coronary syndromes. The appraisal was submitted to the National Institute for Clinical Excellence (NICE) in August 2003 and contributed to the development of the recently published guidelines for the use of clopidogrel in acute...
