Metabolism of the nitrofurans. II. Incubation of Furacin with mammalian tissues.

چکیده
Furacin* (5-nitro-2-furaldehyde semicarbazone) is an antibacterial agent effective against both Gram-negative and Gram-positive organisms (l-3). It has also been studied extensively, along with other nitrofurans, to determine its fate during metabolism in the body (4, 5). Certain of these nitrofurans having chemotherapeutic activity partially withstand metabolic breakdown in the body and are excreted in the urine (4). To extend further the metabolic studies on the nitrofurans, information was sought on the site and mechanism of breakdown of Furacin in the body. Preliminary incubation studies on Furacin with various body tissues are reported in this paper. The studies on intestinal tissue received emphasis because of their significance in connection with concurrent work on the Furacin-xanthine oxidase system (6).

For a better understanding of the mechanism of action of the nitrofurans in the mammalian organism, studies of their metabolism have been initiated. Excretion studies (1) have revealed that only a small part of most ingested nitrofurans appears unchanged in the urine. Bender and Paul (2) in preliminary studies have shown that 5-nitro-2-furaldehyde semicarbazone (Furacin’) undergoes degradation ...
متن کاملMetabolism of the nitrofurans; ultraviolet absorption studies on urinary end-products after oral administration.
The general antibacterial action in vitro of a variety of furan derivatives substituted in the 5 position with a nitro group has been reported by Dodd and Stilhnan (1, 2). That the presence of the nitro group is essential was established by comparison of the activities with those of the corresponding non-nitrated derivatives. All the nitrofurans except those limited by insolubility exhibited a ...

متن کاملEffect of furacin on pyruvate metabolism.
Earlier studies have shown that Furacinl inhibits the formation of acetyl coenzyme A2 from pyruvate anaerobically (1). Acetone powder preparations of pigeon liver and rat testis were used in the early work. The results suggested that Furacin inhibits reactions mediated by pyruvic oxidase; therefore, further studies were continued with more highly purified preparations of this enzyme from pigeon...
متن کاملA glutathione reductase from Escherichia coli.
Earlier studies had demonstrated that a flavoprotein capable of mediating a triphosphopyridine nucleotide (TPN)-linked reduction of Furacin (5-nitro-2-furaldehyde semicarbazone) was present in cell-free extracts of Furacin-susceptible strains of Escherichia coli, but was absent in extracts of resistant strains (I). Dr. Philip E. Hartman, who was then associated with our department, suggested to...
