Microsampling for blood-lead analysis.
نویسندگان
چکیده
We detail the technique, procedure, and equipment required to properly obtain a micro-scale capillary (fingerstick) blood specimen for subsequent determination of its lead content. We divide the microsampling procedure into preparation of sampling equipment and subject, the fingerstick, micro blood collection devices, and transport and storage. The concept of a total-system approach to qualitycontrol for sampling and analysis is presented. For realistic quality control, the sampling component and the analysis component are inseparable. We describe types and sources of errors encountered. Most of the variability in the total system is associated with sampling.

Mouse Pharmacokinetic Study of Ceftriaxone Using MitraTM Microsampling Devices
G. Gu, M. Fang, B. Virasingh, A. Groff, J. Freed, C. Clifton, Y. Li, D. Cheng Alliance Pharma Inc. Purpose Mice are one of the most commonly used animals in drug discovery pharmacokinetic (PK) studies. Guidelines are established for the frequency and amount of blood that can be collected from a single animal. Since the mouse’s body size is limited, parallel blood sampling is generally used wher...
متن کاملDetermination of copper in plasma and serum by use of a microsampling cup in atomic absorption spectrometry.
A microsampling cup procedure is described for rapid, precise determination of copper in plasma. Conventional Delves-cup atomic absorption spectrometry does not yield usable copper signals with blood plasma samples. Analytical sensitivity was improved 11-fold by use of cups fitted with an axial hole, a triangular cup-holding loop, and a smaller-bore absorption tube. The modified procedure yield...
متن کاملPerforated dried blood spots: a novel format for accurate microsampling.
Dried blood spots (DBS) in their current format encounter challenges in bioanalysis using fixed areas, including but not limited to, waste of DBS samples (only a fraction is used for analysis), the need for sample punching leading to concerns of sample carryover, uncertainty for accurate recovery assessments and hematocrit (HCT) effects. Here we describe a novel concept, namely perforated dried...
متن کاملRefinement of Mouse Protocols for the Study of Platelet Thromboembolic Responses In Vivo.
Mouse models of thromboembolism are frequently used to investigate platelet function in vivo and, according to European Union (EU) legislation, must be conducted in the context of replacement, refinement and reduction. We have previously developed a refined real-time mouse model of thromboembolism as an alternative to models of thromboembolic mortality which inflict considerable pain and suffer...
متن کاملBioanalysis of emixustat (ACU-4429) in whole blood collected with volumetric absorptive microsampling by LC-MS/MS.
BACKGROUND A method to quantify emixustat (an investigational drug agent) in human blood collected using volumetric absorptive microsampling (VAMS) could be more practical for sample collection at sites with limited facilities for processing and storage of plasma. METHODS A LC-MS/MS method was developed and evaluated for accuracy and precision, linearity, carryover, selectivity, recovery, mat...
