Immunogold Localization of Tobacco Rattle Virus Particles within Paratrichodorus anemones.

چکیده
Unequivocal evidence of the viral nature of virus-like particles observed at the specific site of retention of tobacco rattle virus (TRV) in Paratrichodorus and Trichodorus nematodes has not previously been available. A new staining technique using safranin-O, which does not affect viral antigenicity, was used with an antiserum raised against the coat protein of TRV and prepared for use with immunogold labelling. Application of this method enabled the occurrence and localization of particles of TRV to be confirmed in the pharynx of the natural vector of the virus, Paratrichodorus anemones, and provided unequivocal evidence that the particles observed were TRV particles. The TRV particles were observed attached only to the cuticle lining the posterior tract of the pharyngeal lumen of the vector. Therefore, the specific site of retention of TRV particles in P. anemones is apparently more localized than reported to occur in other vector trichodorid species.

Eliminating Tobacco Rattle Virus from Viruliferous Paratrichodorus allius and Establishing a New Virus-Vector Combination.
A reliable method to eliminate tobacco rattle virus (TRV) from viruliferous Paratrichodorus allius populations was developed. This virus is vectored by P. allius in the Pacific Northwest and causes corky ringspot disease (CRS) of potato. The viruliferous nematodes that were reared on 'Vernema' alfalfa or '770' scotch spearmint for at least 3 months did not transmit TRV to 'Samsun NN' tobacco, a...
متن کاملThe use of collagenase to improve the detection of plant viruses in vector nematodes by RT-PCR.
Tomato ringspot virus (ToRSV), Tobacco ringspot virus (TRSV) and Tobacco rattle virus (TRV) are transmitted to healthy plants by viruliferous nematodes in the soil. A method was developed for extraction of genomic viral RNA from virus particles carried within nematodes and a sensitive nested RT-PCR detection assay. The procedure has been adapted to microscale for handling multiple samples. This...
متن کاملSimilarities in the genome organization of tobacco rattle virus and pea early-browning virus isolates that are transmitted by the same vector nematode.
Although sequence data have been obtained for several tobravirus isolates, only two of these isolates are nematode-transmissible. Tobacco rattle virus (TRV) PpK20 is transmitted by Paratrichodorus pachydermus, whereas pea early-browning virus (PEBV) TpA56 is transmitted by Trichodorus primitivus. To clarify whether differences in the genome structure of these isolates are relevant to the specif...
متن کاملThe Interaction between Raspberry Ringspot and Tobacco Rattle Viruses in Doubly Infected Protoplasts
Tobacco mesophyll protoplasts infected with raspberry ringspot virus (RRV) show faint generalized staining when treated with fluorescent antibody to RRV, but many brightly staining granules develop after inoculation with a mixture of RRV and the CAM strain of tobacco rattle virus (TRV-CAM). The phenomenon occurred with three different strains of RRV but not when tobacco mosaic virus, or two oth...

Corky ringspot disease (CRS) of potato (Solanum tuberosum) is caused by the tobacco rattle virus (TRV), which is vectored by stubby-root nematodes, Paratrichodorus spp. and Trichodorus spp., and is a significant threat to potato quality and production in many areas of the western United States. Between 2002 and 2005, fields with a history of CRS were planted to potato and treated with various c...
