Efficacy of a Recombinant Genotype VII Vaccine against Challenge with Velogenic Newcastle Disease Virus

چکیده
Background: Newcastle disease virus (NDV) genotype VII has become the dominant genotype in China. However, NDV genotype II was used to make current commercial NDV vaccines. The mismatch of genotypes between circulating and vaccine strains of viruses may compromise the efficacy of vaccines. Methods: In this study, a current circulating NDV was attenuated by mutations of multiple basic amino acid motif of fusion cleavage site 112RRQKGF117 based on reverse genetic techniques. The recombinant virus was prepared as an inactivated vaccine to test its efficacy and compared with a commercial LaSota NDV vaccine on SPF chickens. Results: All vaccinated chickens survived by the end of the study. By contrast, the unvaccinated chickens were all dead before 5 days post-challenge (DPC). Compared to commercial LaSota vaccine, the experimental inactivated vaccine elicited earlier and higher titer of HI antibodies and had reduced titer and duration of virus shedding after challenge. Conclusion: The experimental inactivated NDV vaccine may work as a promising vaccine candidate to control the disease. NDV genotype II strains [7]. The cross-protection of vaccines to genotype VII virus was seldom explored. In this study, we developed an inactivated NDV genotype VII vaccine based on reverse genetic techniques and compared its efficacy with commercial LaSota vaccine on SPF chickens. Material and methods

Efficacy of inactivated velogenic Newcastle disease virus genotype VII vaccine in broiler chickens
Newcastle disease (ND) causes severe economic losses in poultry production. Despite the intensive vaccination regimes of NDV in Egypt, many outbreaks are being reported. The present study focused on the preparation and evaluation of inactivated velogenic Newcastle disease virus vaccine (genotype VII) isolated from Egyptian broiler chicken during 2015-2016. Fifty-five tissue samples including tr...

A Newcastle disease virus (NDV) isolate designated IBS002 was isolated from a commercial broiler farm in Malaysia. The virus was characterised as a virulent strain based on the multiple basic amino acid motif of the fusion (F) cleavage site (112)RRRKGF(117) and length of the C-terminus extension of the hemagglutinin-neuraminidase (HN) gene. Furthermore, IBS002 was classified as a velogenic NDV ...
متن کاملThe Combined Use of rHVT-H5 and rHVT-F Vector Vaccines in the Hatchery Enhances Immunity against Highly Pathogenic Avian Influenza H5N1 and Velogenic Newcastle Disease Viral Infections in Commercial Chickens
Highly pathogenic avian influenza H5N1 and Newcastle disease viral infections cause severe illness in chickens and vaccination is a strategic tool of controlling these diseases. Hence, this study was conducted to evaluate the efficacy of using both recombinant herpesvirus of turkey (rHVT-H5 and rHVT-F) vector vaccines at day-old, in the hatchery, under field conditions. Vaccinated chickens were...

Newcastle disease virus (NDV) is a prototype member of avian paramyxovirus serotype 1 (APMV-1), which causes severe and contagious disease in the commercial poultry and wild birds. In this study, we report the results of phylogenetic analyses of recent NDVs isolated from Markazi province of Iran during a recent outbreak in the commercial broiler with respiratory signs and digestive system lesio...
متن کاملF gene recombination between genotype II and VII Newcastle disease virus.
A velogenic Newcastle disease virus (NDV) strain, designated as SRZ03, was isolated from an egg layer flock with NDV vaccine immunization failure in China in 2003. Recombination was found in the F gene of SRZ03. Complete genome sequences analysis indicated that the N-terminal of SRZ03 F gene originated from a genotype II NDV strain, whereas the C-terminal of F gene and the rest of the genes ori...
