Thermosensitization by Sulfhydryl Compounds of Exponentially Growing Chinese Hamster Cells1

چکیده
The effect of various sulfhydryl compounds on the survival of exponentially growing monolayer cultures of Chinese ham ster cells (HA1 ) heated to temperatures of 37-43° was exam ined. Concentrations of cysteamine which were nontoxic or minimally toxic at room temperature or 37° became increas ingly toxic at elevated temperatures, greatly potentiating the killing produced by heat alone in the absence of cysteamine. This enhancement of hyperthermia-induced cell killing in creased with increasing cysteamine concentration, increasing duration of cysteamine exposure, and increasing temperature. Studies with synchronized Chinese hamster cells heated at 43° for 1 hr in the presence of 16 rnw cysteamine demonstrated that the potentiation of heat killing occurred in all phases of the cell cycle. Similarly, enhancement of hyperthermia-induced cell killing was seen for asynchronous cells exposed to 2-aminoethylisothiourium bromide and cysteine, but the magnitude of the effect differed for the various sulfhydryl compounds.

The findin«that 7r,8f-dihydroxy-9,10f-oxy-7,8,9,10-tetrahydrobenzo(a)pyrene (BPDE-I) is stabilized against hydrolysis by binding to cellular membranes suggested that nucleophilic compounds which would colocalize with BPDE-I in membranes might inhibit the deleterious biological effects of BPDE-I. We have explored the possibility that hydrophobic, sulfhydryl-containing compounds might provide su...
متن کاملChemical modification of the neutral amino acid transport system L of Chinese hamster ovary cells with p-chloromercuribenzene sulfonate.
Branched-chain and aromatic neutral amino acids enter mammalian cells predominantly through a Na(+)-independent transport agency called System L. The sulfhydryl specific reagent p-chloromercuribenzene sulfonate (pCMBS) has been shown to be a potent inactivator of System L transport activity in Chinese hamster ovary cells, however, inactivation by pCMBS can be prevented by the presence of System...

The relationship between spermine-induced thermosensitization and modulation in the cellular redox state as measured by glutathione levels was studied using Chinese hamster V79 cells. Marked cellular glutathione depletion was observed for cells treated with exogenous 1 mM spermine at 37 degrees C or 43 degrees C. Glutathione depletion and thermal sensitization by spermine were found to be cell ...
