Inhibition of nucleic acid synthesis by folic acid antagonists.

چکیده
Woolley and Pringle (13) have recently reported the formation of 4-amino-5-carboxamidoimidazole during growth of Escherichia coli in the presence of 4-aminopteroylglutainic acid (aminopterin). Some of us have observed that excessive doses of folie acid speed up the leukemic process in mice and that folie acid will reverse the well known antileukemic action of aminopterin (11). This reversal of the anti-leukemic action of folie acid antagonists by means of large doses of folie acid had previously been reported by Burchenal (4) and Law (7). In the face of the above information, the sugges tion that formyl folie acid might be involved in in-

Amethopterin and related folic antagonists inhibit the reduction of folic acid to tetrahydro folic acid (2). The clinical effects of these drugs are thought to be related in turn to the role of tetrahydro folic acid in single-carbon transfer reactions during nucleic acid synthesis (3). Recently, pteridines such as folic acid have been implicated in another area of metabolism, the conversion of ...

that folic acid antagonists provide temporary palliation in certain cases of acute leukemia in children (7) and that several compounds of this class increase the survival time of mice with transplanted leukemia Ak 4 (4, 10, 11). However, eventual failure in the treatment of leukemia by chemotherapy with such compounds occurs consistently. The mechanism involved in the development of drug resist...

Enzyme induction in Escherichia coli is stimulated by certain 5-amino-2,4-bis(substitutedamino)pyrimidines (Kunkee, 1960a). Other 2,4diaminopyrimidines have been reported to be antagonists of folic acid (Hitchings et al., 1952; Doctor, 1956). Since there are indications that the stimulation of enzyme induction involves nucleic acid metabolism (Kunkee, 1960b) and since folic acid is involved in ...

Inhibition of the growth of Lactobadlus casei with folic acid as the nutrilite is a general property of 2,4-diaminopyrimidines, 2-aminopyrimidines, and condensed systems containing these groupings (1). These compounds comprise a spectrum of inhibitors. At one end lie the simpler diaminopyrimidines which are competitive antagonists of folic acid (1, 2). The center may be represented by 2-aminopu...

Considering the high water requirement of grass during growth stages and the limitation of irrigation sources, it is important to find a suitable way to reduce the water requirement of the lawns as cover plants used in the Landscape. The purpose of this study was to investigate the effect of 2,4,5-T on the induction of callus from the hypocotyl explant in Kentucky bluegrass, induce in vitro dro...
