Agrobacterium-mediated transformation of Brassica rapa with a bacterial isochorismate synthase (ICS) gene

چکیده
The entC gene from Escherichia coli which codes for isochorismate synthase (ICS) was introduced into Brassica rapa varieties i.e. var. rapa cv. Raapstelen and ssp. oleifera. Transgenic plants of ssp. oleifera carrying the ICS gene have been developed via Agrobacterium tumefaciens-mediated transformation. This method involved the use of hypocotyl explants co-cultivated with disarmed Agrobacterium strain LBA4404, which on the T-DNA region carries CaMV 35S promoter driven genes ssentC, consisting of the entC coding region fused to the coding region of the plastid targeting sequence of tobacco ribulose-bisphospate carboxylase, hptII, encoding hygromycin phosphotransferase, and a gene for sGFP. Prior to this, several important parameters were examined for their influence on the transformation efficiency. The presence of the transgenes in the genome of the nine months old greenhouse-grown transgenic plants was confirmed by PCR, whereas the expression of the entC gene was revealed by RT-PCR. The results indicated that the exogenous gene was successfully integrated into the genome and expressed in transgenic B. rapa plants.

Salicylic acid accumulation in transgenic Brassica rapa transformed with a gene encoding bacterial isochorismate synthase
The accumulation of salicylic acid (SA) in Brassica rapa ssp. oleifera transformed with a bacterial isochorismate synthase (ICS) gene was examined in the old and young leaves of the primary transformants through high performance liquid chromatography analysis. The level of SA and SA glucoside (SAG) in the transformed plants varied between the individual plants but the SA contents of the leaves ...

The biosynthetic pathways of saturated and unsaturated fatty acids consist of many steps controlled by various enzymes. One of the methods for improving oil quality is to change the fatty acid profile through genetic manipulation which requires isolation and characterization of the genes and other cis-acting elements, such as the promoter, involved in fatty acid biosynthesis. b-ketoacyl-CoA syn...
متن کاملA simple and efficient Agrobacteriumtumefaciens-mediated plant transformation of Brassica rapa ssp. pekinensis
The present study aims to investigate the numerous factors influencing Agrobacterium tumefaciens-mediated genetic transformation of Chinese cabbage (Brassica rapa ssp. pekinensis). Factors affecting transformation efficiency, such as age of explants, Agrobacterium concentration, and effect of acetosyringone, pre-cultivation, infection and co-cultivation time of Agrobacterium were examined. The ...
متن کاملSalicylic acid and systemic acquired resistance play a role in attenuating crown gall disease caused by Agrobacterium tumefaciens.
We investigated the effects of salicylic acid (SA) and systemic acquired resistance (SAR) on crown gall disease caused by Agrobacterium tumefaciens. Nicotiana benthamiana plants treated with SA showed decreased susceptibility to Agrobacterium infection. Exogenous application of SA to Agrobacterium cultures decreased its growth, virulence, and attachment to plant cells. Using Agrobacterium whole...
متن کاملOptimization of Agrobacterium-mediated transformation in oyster mushroom (Pleurotus ostreatus) by vector containing human pro-insulin gene
Transferring foreign genes into mushroom mediated by Agrobacterium tumefaciens is a standard technique in genetic engineering. Recombinant human insulin has been greatly used in the treatment of type I diabetes. The production of edible mushroom derived insulin should facilitate oral delivery. In this study we used the Agrobacterium tumefaciens mediated transformation method for the transfer an...
