چکیده

When Detroit cells are infected with poliovirus in the presence of 5/~g./ml. of actinomycin, cell-controlled RNA synthesis is blocked and 2-[14C]uridine is incorporated only into poliovirus-specific RNA. The addition of polyinosinic-polycytidylic acid (5/%/ml.) under such conditions results in the inhibition of synthesis of 35s intracellular, infectious poliovirus RNA. A high molecular weight fraction of poliovirus-specific RNA migrating on polyacrylamide gels, as would be expected of the double-stranded, replicative form of poliovirus RNA, is synthesized to the same extent as in the absence of interferon inducer. This RNA was digested with RNase and DNase and was resistant to the enzymes, confirming that the compound was replicative form poliovirus RNA. A low molecular weight, virus-induced RNA of 4s and unknown function is also synthesized independently of polyinosinic-polycytidylic acid. The significance of the differential inhibition in the synthesis of the compounds presumed to be poliovirus RNA and replicative form poliovirus RNA, and the fact that the capacity of polyinosinic-polycytidylic acid to inhibit the synthesis of infectious RNA is insensitive to actinomycin, is discussed.

مقایسه تأثیر Tumor Necrosis Factor-α، Monocyte Conditioned Medium و polyinosinic-polycytidylic acid در بلوغ سلول‌های دندریتیک

Abstract Background and purpose: Different laboratories have considered different maturation factors for producing human dendritic cells (DC) from peripheral blood monocytes. In this study, we comprehensively investigated the effect of adding poly (I:C) to standard maturation stimulus, i.e. MCM and TNF-α on the induction of T cell response. Materials and methods: Peripheral blood monocytes ...

Protection from RNA and DNA viruses by IL-32.

Several studies have documented a proinflammatory role for IL-32, which induces IL-1α, IL-1β, IL-6, TNF, and chemokines via NF-κB, p38MAPK, and AP-1. However, IL-32 also participates in the responses to infection with viruses such as HIV-1 and influenza. In this study, we explored these antiviral properties of IL-32. Vital staining assays demonstrated that low concentrations (5-10 ng/ml) of rIL...

Distinct CpG DNA and polyinosinic-polycytidylic acid double-stranded RNA, respectively, stimulate CD11c- type 2 dendritic cell precursors and CD11c+ dendritic cells to produce type I IFN.

Two classes of nucleic acids, bacterial DNA containing unmethylated CpG motifs and dsRNA in viruses, induce the production of type I IFN that contributes to the immunostimulatory effects of these microbial molecules. Thus, it is important to determine which cells produce type I IFN in response to CpG DNA and dsRNA. CD4(+)CD11c(-) type 2 dendritic cell precursors (pre-DC2) were identified as the...

The role of IFN regulatory factor-3 in the cytotoxic activity of NS-9, a polyinosinic-polycytidylic acid/cationic liposome complex, against tumor cells.

NS-9 is a complex of polyinosinic-polycytidylic acid and a novel cationic liposome, LIC-101. The complex has strong cytotoxic activity against tumor cells derived from epithelial or fibroblastic cells. We have investigated the mechanism of the cytotoxic activity of NS-9 using knockdown cells in which the expression of proteins of interest was inhibited by RNA interference. NS-9 showed strong cy...
