A novel sialic acid and fucose-containing disaccharide isolated from the jelly coat of sea urchin eggs.

چکیده
In our previous investigations, two sialic acids were isolated from the jelly coat of the eggs of the sea urchin Pseudocentrotus depepr~ssl~s; one was N-glycolylneuraminic arid, the other was W-acctoglycolyl-4-methyl-4,9-dideosyncuraminic acid (2). The presence of additional sialic acid compounds was detected in the mixt’urc resulting from mild acid hydrolysis of the starting material (1). Isolation and purification of one of these perceptible disaccharides and its eventual characterization as a sialic acid and fucose containing compound are the subjects of this present study.

Isolation of VLs from Unfertilized Eggs
The vitelline layers (VLs) of unfertilized sea urchin eggs were isolated by homogenization in a hypotonic medium containing Triton X-100 and EDTA. The surface topography of the VL is not changed by isolation. The thickness of the isolated VLs (300-400/~) is greater than that reported for VLs on intact eggs (100-200 .~). Sperm adhere to the isolated VLs. When both internal and external VL surfac...
متن کاملIsolation and identification of two sialic acids from the jelly coat of sea urchin eggs.
Two sialic acids were isolated from the jelly coat of the eggs of the sea urchin Pseudocentrotus depressus. Both were subjected to chemical assay, thin layer chromatography, infrared absorption study, and enzymatic treatment. The results indicate that one of the sialic acids is identical with N-glycolylneuraminic acid. The other sialic acid is a completely new type and was deduced to be N-aceto...

متن کاملAgglutination of jelly coat and cortical granule components and the block to polyspermy in the amphibian Xenopus laevis.
A block to polyspermy in amphibians is established at fertilization by the conversion of the vitelline envelope to the fertilization envelope. In Xenopus laevis a major ultrastructural change in the envelope at fertilization is the appearance of an electron-dense layer, termed the F layer, between the envelope and the inner-most jelly coat layer, J(1). The F layer is derived, at least in part, ...
متن کاملEgg sialoglycans increase intracellular pH and potentiate the acrosome reaction of sea urchin sperm.
Sea urchin egg jelly (EJ) triggers sperm acrosome reaction (AR), an exocytotic event required for membrane fusion of the gametes. Purified fucose sulfate polymer (FSP) in EJ is one inducer of the AR. Binding of FSP to its receptor regulates opening of two distinct calcium channels and also elevates intracellular pH (pH(i)). EJ also contains sialic acid-rich glycans (sialoglycans (SG)) that were...
