Construction of Genetic Linkage Map and QTL Analysis for Fiber Traits in Diploid Cotton (Gossypium arboreum x Gossypium herbaceum)

چکیده
Diploid A genome cottons are grown for their natural fiber in drought prone areas of Asia. Although they possess inferior fiber qualities to cultivated Upland and Pima cotton, their value lies in their inherent resistance to pests and diseases. Molecular linkage maps provide essential tools for plant genetic research, facilitating quantitative trait locus (QTL) mapping, marker-assisted selection and map based cloning. In the present study, we used SSR, AFLP, and TRAP markers to construct a linkage map using 94 F2 diploid individuals derived from a cross between G. arboreum x G. herbaceum. A total of 606 polymorphic markers were used to construct 37 linkage groups covering a total of 1109cM with an average distance of 7.92cM between two loci. Discriminant analysis identified three markers each associated for petal color and seed fuzziness, and four markers for petal spot. For quantitative traits, a total of 19 QTL were identified for five fiber traits using composite interval mapping. The identified QTL’s such as qFL4-1, qFS4-2, qELO11 and qSI2-1 were found to be significantly linked to fiber length, strength, elongation and seed index, respectively. This work could lead to more such attempts in deploying markers onto the A genome, thus guiding QTL mapping in tetraploid cottons.

COMPARISON OF DNA MARKER ORDER AND FLORAL TRAIT EVOLUTION AMONG DIPLOID AND TETRAPLOID GENOMES OF GOSSYPIUM by APARNA DESAI
Scientific information on the A genome species of Gossypium is limited due to their decline as crop species and low levels of marker polymorphism. As the A subgenome of the allotetraploid species of Gossypium is contributed by A genome progenitors, studies on the A genome will help us understand the evolution of polyploids. We compared DNA marker order along the chromosomes of A, D and At genom...
متن کاملIndependent Domestication of Two Old World Cotton Species
Domesticated cotton species provide raw material for the majority of the world's textile industry. Two independent domestication events have been identified in allopolyploid cotton, one in Upland cotton (Gossypium hirsutum L.) and the other to Egyptian cotton (Gossypium barbadense L.). However, two diploid cotton species, Gossypium arboreum L. and Gossypium herbaceum L., have been cultivated fo...
متن کاملHigh-density genetic linkage map construction by F2 populations and QTL analysis of early-maturity traits in upland cotton (Gossypium hirsutum L.)
Due to China's rapidly increasing population, the total arable land area has dramatically decreased; as a consequence, the competition for farming land allocated for grain and cotton production has become fierce. Therefore, to overcome the existing contradiction between cotton grain and fiber production and the limited farming land, development of early-maturing cultivars is necessary. In this ...
متن کاملInsights into the Evolution of Cotton Diploids and Polyploids from Whole-Genome Re-sequencing
Understanding the composition, evolution, and function of the Gossypium hirsutum (cotton) genome is complicated by the joint presence of two genomes in its nucleus (AT and DT genomes). These two genomes were derived from progenitor A-genome and D-genome diploids involved in ancestral allopolyploidization. To better understand the allopolyploid genome, we re-sequenced the genomes of extant diplo...
متن کاملEffects Of Colchicine Treatments On Chromosome Doubling In Three Diploid Cotton Species
Colchicine has been used to induce chromosome doubling in plant species. Diploid cotton species have inherited desirable genes that are resistant to biotic and abiotic stresses and also possesses useful fiber quality traits, such as higher fiber length and strength. To transfer these desirable traits to the most extensively cultivated tetraploid cotton species through hybridization, it is a pre...
