چکیده

The molecular basis of gd T cell receptor (TCR) recognition is poorly understood. Here, we analyze the TCR sequences of a natural gd T cell population specific for the major histocompatibility complex class Ib molecule T22. We find that T22 recognition correlates strongly with a somatically recombined TCRd complementarity-determining region 3 (CDR3) motif derived from germ line– encoded residues. Sequence diversity around these residues modulates TCR ligand-binding affinities, whereas V gene usage correlates mainly with tissue origin. These results show how an antigen-specific gd TCR repertoire can be generated at a high frequency and suggest that gd T cells recognize a limited number of antigens.

Lymphocytes: Gamma Delta

Antigen recognition by B and T lymphocytes is achieved by highly diverse heterodimeric receptors, the Band T-cell receptors (TCRs). T lymphocytes can be subdivided into two mutually exclusive subpopulations carrying TCR chains encoded by either a and b or g and d gene loci. These T-cell subsets are classically referred to as ‘ab’ and ‘gd’ T cells, respectively. Like other antigen receptors, the...

Determinants on T Cell Factors Coded

Thymus-derived(T) lymphocytes have been shown to display antigen specificity. The antigen-recognition units on T lymphocytes, in contrast to those on B lymphocytes, do not appear to be of the conventional immunoglobul in (Ig) type. Recent studies from various laboratories (1-5) have demonstrated that the antigen-specific T cell factors derived from helper or suppressor T cells possess the ant i...

Diversity of T Cell Recognition of Hemagglutin

An essential feature of the immune system is its ability to recognize and respond to a diverse spectrum of foreign antigens encountered in the environment. Two kinds of antigen specific receptors are used, the Ig receptor on B cells and the antigen-specific receptor on T cells. In contrast to Ig, which binds antigen directly, the T cell antigen receptor binds antigen only in association with se...

NK cells engineered to express a GD2-specific antigen receptor display built-in ADCC-like activity against tumour cells of neuroectodermal origin

Treatment of high-risk neuroblastoma (NB) represents a major challenge in paediatric oncology. Alternative therapeutic strategies include antibodies targeting the disialoganglioside GD(2) , which is expressed at high levels on NB cells, and infusion of donor-derived natural killer (NK) cells. To combine specific antibody-mediated recognition of NB cells with the potent cytotoxic activity of NK ...
